Cephalotaxus sinensis(C.sinensis)is an endemic tree species in China,and it can be used for medicine and has a high medicinal value.However,the growth of C.sinensis is slow and lack of natural resources,which is not conducive to the sustainable development of C.sinensis.Therefore,it is a great significance to exploit the high added value utilization potential of C.sinensis,protective development and utilization of C.sinensis resources,and to reveal the potential utilization value of C.sinensis.In this study,the chemical components and contents of extracts from different parts of C.sinensis(leaves,branches,barks,woods and roots)were systematically analyzed by Fourier transform infrared spectroscopy(FTIR),Gas chromatography-mass spectrometry(GC-MS),Quadrupole time-of-flight-liquid chromate graphy-mass spectrometry(QTOF-LC-MS).The results proved that the active compounds had anticancer effect.The chemical components and contents of volatile substances in various parts of C.sinensis were analyzed by Thermal desorber-gas chromatography-mass spectrometry(TD-GC-MS),and the cytotoxicity of C.sinensis to HepG2,leukemia cell K562 and gastric cancer cell sgc-7901 was studied by MTT method.The specific results are as follows:(1)The infrared spectra of ethanol,benzene/ethanol(1:1)and methanol extracts from the leaves,branches,barks,woods and roots of C.sinensis were analyzed.It was found that the absorption peaks of the extracts were mainly concentrated in the wavenumber range of 3600-3200 cm-1,3000-2850 cm-1 and 1760-1000 cm-1,the chemical bonds are:C-H,O-H,C=O,C-O-C,P-O-C,C-O,C=C,C-C.The main chemical components may be as follows:alcohols,phenols,carboxylic acids,alkanes,alkenes,esters,aldehydes,aromatics,amides,acetyl.(2)The results of GC-MS analysis showed that there were 121 compounds in C.sinensis,of which 48 were the most active compounds with beneficial characteristics.The distribution of active compounds in different parts of C.sinensis was as follows:18 in leaves,13 in branches,18 in barks,17 in woods and 16 in roots.Among them,cephalotaxine as an effective anticancer medicine,the content of cephalotaxine extracted from various parts is as follows:3.41%in leaves,6.13%in branches,32.73%in woods and 23.56%in roots.(3)The QTOF-LC-MS detection of ethanol and distilled water extracts of various parts of the Cephalotaxus sinensis showed that there are as many as 204 compounds in the crude torreya,114 of which have beneficial characteristics.The distribution of active compounds in each part is as follows:There are 88 kinds of leaves,84 kinds of branches,114 kinds of bark,82 kinds of trunks,and 96 kinds of roots.The main discoveries are riboflavin,which has the effects of clearing heat and relieving heat,raising Yang and cooling blood,and hemostasis,genipin,which has anti-cancer and hepatoprotective effects,and scopolamine,which has therapeutic effects on motion sickness and Parkinson’s disease.(4)TD-GC-MS analysis showed that there were 244 compounds in C.sinensis.Which has 71 species of leaves,77 species of branches,83 species of barks,83 species of woods and 85 species of roots.It contains squalene,which can improve liver function and tissue activity,isosorbide nitrate,which can be used as a quick acting and long-acting nitrate anti-angina medicine,and sickle leaf celery alcohol,which has neuroprotective activity and is used in the treatment of Alzheimer’s disease.(5)MTT analysis showed that the ethanol extracts of C.sinensis had inhibitory effect on HepG2 cells,K562 cells and Sgc-7901 cells.The lethal rate of ethanol extracts from C.sinensis woods to Sgc-7901 cells was 93.83%.In summary,the C.sinensis extract is rich in chemical molecular resources and rich in biologically active ingredients,especially the ethanol extract of C.sinensis contains a large amount of anticancer active substances such as harringtonine,morin,rhein,luteolin,etc.,And has significant cytotoxicity.Therefore,C.sinensis has a good prospect for the development of new anti-cancer drugs.At the same time,it provides theoretical basis and technical support for tapping the potential of high-value utilization of the resources of C.sinensis.
