| The aim of this study was to investigate the effects of 15 mg/kg pioglitazone hydrochloride(PGZ),400 mg/kg L-carnosine(LC)and 400 mg/kg resveratrol(RES)on the growth performance,slaughter performance,serum biochemical indicators,muscle antioxidant properties,intramuscular fat content and fatty acid composition of yellowfeathered broiler chickens,and preliminary explored its mechanism.In the experiment,750 female yellow-feathered broiler chickens(dwarf-yellow chicken,average weight 1624.11±8.44 g),70 days old(28 days before market),were randomly divided into 6 treatment groups for 28 d,with 5 replicates in each treatment group and 25 chickens per replicate.The control group was fed the basic diet,and the experimental group was fed the basic diet supplemental with 15 mg/kg PGZ,400 mg/kg LC,400 mg/kg RES,15 mg/kg PGZ +400 mg/kg LC and 15 mg/kg PGZ +400 mg/kg RES,respectively.The broiler chickens were allowed access to feed and water ad libitum.The result shows:(1)Compared with the control group,dietary supplemental with PGZ,LC,PGZ+LC or PGZ+RES could significantly increase the average daily weight gain of yellow-feathered broiler chickens and reduce the feed weight gain ratio(P<0.05);Adding RES alone to the diet did not affect the growth performance of broilers(P>0.05);adding PGZ,PGZ + LC or PGZ + RES to the diet could significantly increase the slaughter rate of yellow-feathered broiler chickens and reduce its abdominal fat rate(P<0.05).(2)Dietary supplemental with PGZ,PGZ+LC or PGZ+RES could significantly reduce the serum levels of low-density lipoprotein cholesterol(LDL-C)and cholesterol(CHO)of yellow-feathered broiler chickens,while increasing the serum level of high-density lipoprotein cholesterol(HDL-C)(P<0.05).(3)Dietary supplemental with LC,RES,PGZ+LC or PGZ+RES could significantly reduce the drip loss and cooking loss of the thigh and breast muscles(P<0.05);diet supplementation with LC or PGZ+LC could significantly increase the redness(a*)value of the thigh muscle(P<0.05);diet supplementation with PGZ,PGZ+LC or PGZ+RES could significantly increase the intramuscular fat(IMF)content in the breast and thigh muscles(P<0.05),and increase the percentage of C18: 1n9,C20: 3n3,C18: 3n6 and monounsaturated fatty acids(MUFA)in the thigh muscles(P<0.05).(4)Dietary supplemental with LC,RES,PGZ+LC or PGZ+RES could significantly increase the activity of glutathione peroxidase(GSH-Px)and superoxide dismutase(SOD)(P<0.05),and reduce the content of malondialdehyde(MDA)and reactive oxygen species(ROS)in the thigh muscle(P<0.05).(5)Dietary supplemental with PGZ,PGZ+RES or PGZ+LC could significantly increase the mRNA abundance of lipogenesis-related genes in thigh muscles of yellow-feathered broiler chickens,such as peroxisome proliferator-activated receptor γ(PPARγ),peroxisomes receptor γ coactivator 1α(PGC-1α)and fatty acid binding protein 3(FABP3)(P<0.05);diet supplementation of LC,RES,PGZ+RES or PGZ+LC could significantly increase the mRNA abundance of antioxidation-related genes in the thigh muscle,such as nuclear transcription factor red-2 related factor 2(NRF2)and superoxide dismutase 1(SOD1)(P<0.05).Conclusion: Diet supplemental with 15 mg/kg PGZ alone to the diet could significantly improve the growth performance,slaughter performance,IMF and MUFA content of yellowfeathered broiler chickens;diet supplemental with 400 mg/kg LC or 400 mg/kg RES alone could significantly reduce the drip loss and cooking loss of breast muscle and thigh muscle,and improve the antioxidant capacity of thigh muscle;diet supplemental with 15 mg/kg PGZ+400 mg/g LC or 15 mg/kg PGZ +400 mg/kg RES could significantly improve the growth performance and slaughter performance of yellow-feathered broiler chickens,and improve the muscle quality and antioxidant ability of breast and thigh muscles,showing an obvious additive effect;this may be related to the synergistic upregulation of the mRNA abundance of PPARγ,PGC-1α,FABP3,NRF2 and SOD1 mRNA in muscle when dietary supplemental with PGZ+LC or PGZ+RES. |
