Forsythia suspensa(Thunb.)Vahl.is a traditional Chinese herbal medicine for clearing away heat and toxic material,with good anti-inflammatory effects.Forsythoside A,phillyrin and phillygenin are potential anti-inflammatory active composition in forsythia suspensa.Near-infrared spectroscopy technology is one research focus for the quality control of Chinese herbal medicine recently.The basis of the efficacy of Chinese herbal medicine is the content of effective composition.This article mainly uses near-infrared spectroscopy to analyze the quality of forsythia suspensa,and also studies the anti-inflammatory activity of the water abstract of forsythia suspensa.The experiment is divided into the following four aspects:The first part is the determination of the contents of forsythiaside A,phillyrin and phillygenin on the main anti-inflammatory active composition in forsythia suspensa by the HPLC.The content of forsythiaside A and phillyrin are different in August and October.The data of forsythiaside A,forsythin,and forsythiaside not only provide data support for the qualitative and quantitative analysis of forsythia by near-infrared spectroscopy,but also provide a theoretical basis for the study of anti-inflammatory active composition of forsythia suspensa.The second part is the application of near infrared spectroscopy in the qualitative analysis of Forsythia.The discriminant analysis method was used to establish the qualitative analysis model of"Qing Qiao"and"Lao Qiao",and the calibration set and prediction set samples were located in the corresponding quadrants(P<0.05).The results show that all samples can be accurately classified and the analysis results are highly accurate.The third part uses partial least squares(PLS)to establish the quantitative analysis models of forsythiaside A,forsythin,and forsythiaside in Forsythia.The RPD value of the established quantitative analysis model was within a good range,and the prediction accuracy is igh,could be used for rapid determination of forsythin A,forsythin and forsythin.In the fourth part,LPS was used to stimulate RAW264.7 cells to establish an inflammation model.By detecting the apoptosis rate,NLRP3,LC3Ⅱ/LC3Ⅰ,PI3K and AKT and other indicators,we observed the effects of different concentrations of FWA on the RAW264.7 cells inflammation model.The effect of FWA on autophagy and PI3K-AKT-m TOR pathway.The results showed that the water extract of Forsythia can inhibit the apoptosis of RAW264.7 cells,and promote cell proliferation at 20,2-1,2-2and 2-3mg/m L(P<0.05);FWA is the most suitable The drug concentration is 0.1 mg/m L,0.2 mg/m L and 0.4 mg/m L;0.4mg/m L FWA could inhibit the release of inflammatory proteins such as NLRP3(P<0.05),reduce the ratio of LC3Ⅱ/LC3Ⅰ,and promote the production of PI3K and AKT proteins.It was further suggested that the potential anti-inflammatory mechanism of the water extract of Forsythia may inhibit autophagy by activating the PI3K/AKT pathway.
