| Cassava(Manihot esculenta Crantz)is an important cash crop and food source growing in Africa,Asia,and tropical America because of its high starch content in storage root;about 750 million people depend on cassava as food.Tubulin/Fts Z family is a group of conserved GTP-binding proteins,As the main component of cytoskeleton,this family is involved in cell division,cell movement,intracellular transport and cell shape control,and is closely related to plant growth and development.Ftsz2-1 belongs to Tubulin superfamily and plays a key role in chloroplast division.In this study,the tubulin family of cassava was screened by bioinformatics method;The biochemical characteristics,predictive subcellular localization,phylogenetic tree analysis,exon-intron structure,conserved motifs,chromosome localization and gene replication of tubulins were analyzed;the expression pattern of FtsZ2-1 under different stresses were analyzed by q RT-PCR;The effect of FtsZ2-1 overexpression on starch accumulation in cassava was analyzed,and the molecular mechanism of its effect on starch accumulation was analyzed by transcriptome sequencing,which laid a foundation for improving the quality of cassava starch.The main results are as follows:(1)A total of 23 cassava Tubulins(Me Tubulins)were identified and divided into four main groups according to their type,phylogenetic characteristics and exon-intron structure.FtsZ2-1(Me Tubulin10)was divided into group IV.(2)Me Tubulins is unevenly distributed on chromosomes.(3)Fragment replication is an important driving force for the expansion of tapioca Tubulin gene family.(4)The expression of Tubulin family members in different tissues showed that most of them were expressed in mid-vein,petiole and stem.(5)The results of q RT-PCR of FtsZ2-1 gene under hormone and abiotic stress showed that in-dole-3-acetic acid(IAA),methyl jasmonate(Me JA),gibberellin A3(GA3),polyethylene glycol(PEG)and salt(Na Cl)could induce the expression of FtsZ2-1 gene,while it showed good stability under the induction of abscisic acid(ABA).(6)The starch content of FtsZ2-1 transgenic cassava increased by 12~17%.Transcriptome analysis showed that carbon fixation of photosynthetic organisms,temporary starch formation by gluconeogenesis,sucrose and starch biosynthesis pathway were affected. |
