Mangifera(Mangifera indic)is an evergreen fruit tree of the Anacardiaceae family.It is a famous tropical fruit and has important economic value.A series of physiological and biochemical changes during mango fruit development and postharvest storage,such as color change,sugar accumulation and stress response under low temperature stress,have an important impact on its commercial properties.These biological processes are related to MYB,bHLH,WD40 and RAV.Transcription factors are closely related.This research is based on a global comparison of the whole mango genome,using bioinformatics methods to analyze its physical and chemical properties,conserved domains,conserved motifs,gene structure,phylogeny,homology,homeopathic elements,protein interactions,etc.Perform analysis and then combine the published transcriptome data(involving the mature leaves,mature bark,seeds,roots,peel,pulp and flowers of ’Alphonso’ mango fruit;the peel and pulp of ’Sensation’ mango from green to full ripening;Three varieties of ’Sensation’Hongyu’,’Guire-82’ green ripening stage and ripening stage;the samples are ’Kent’ mango peels under different storage time and low temperature;the samples are hot water treatment and The control ’Ataulfo’ mango peel samples at the green and mature stages;the samples are the ’Tainong-1’ and ’Renong-1’ mango flesh samples at 30 days after flowering and at full maturity.)and q RT-PCR Experiment reached(samples are the peel and pulp of the red variety ’Guifei’,the yellow variety ’Jinhuanggao’,the green variety ’Qingmang’,the high-sugar variety ’Tainong-1’,and the low-sugar variety ’Renong-1’)for expression analysis,Screen out genes with tissue preference,color-related and post-harvest storage-related genes.The main findings are as follows:1.A total of 44 R2R3 MYB family members,103 bHLH family members,108 WD40 family members and 6 RAV family members were screened.2.Mi MYBs are distributed on 18 chromosomes.It is divided into 10 subgroups,and there are similar conserved motifs,conserved domain positions and gene structures among the same subgroups.There are 25 pairs of collinearity among MYB members.It is speculated that the expansion of the MYB gene family in mangosteen may be closely related to the large chromosome replication event.The value of Ka/Ks of the collinearity gene is far less than 1,indicating that the MYB gene of mango fruit may be mainly subjected to purification and selection during the evolution process.The expression of MYBs in different tissues has a preference.Mi MYB06,Mi MYB20 and Mi MYB27 may be involved in the regulation of anthocyanin synthesis in red mango peel.Mi MYB15 may be of great significance for maintaining the necessary life activities of mango cells.3.Genomic mapping analysis revealed that the bHLHs genes of mangosteen were unevenly distributed on 20 chromosomes and one unidentified Scaffold23.Phylogenetic analysis shows that it can be divided into 19 subgroups.The same subgroup has similar conserved motifs,conserved domain positions and gene structures.Collinearity and selective pressure analysis revealed that there are 48 pairs of collinearity among the bHLH members of mangosteen,and 3 sets of tandem repeats.The number of homologous pairings between mangosteen and Arabidopsis bHLH is 39,and that mangosteen and orange bHLH are homologous.The number of pairs is 69.It is speculated that the expansion of the bHLH gene family in mangosteen may be closely related to the tandem duplication and large chromosome duplication events.The selection pressure analysis found that the Ka/Ks value of most collinearity genes was far less than 1,indicating that the mango bHLH gene may be mainly subjected to purification selection to remove harmful mutations during the evolution process.Protein interaction analysis revealed that multiple mango bHLH proteins have an interaction relationship,which involves processes such as plant development,flower formation,and mineral element absorption.Expression analysis found that the expression of mango bHLHs in different tissues is biased,and may be involved in the regulation of multiple biological pathways of mangos.Three bHLH genes(MibHLH027,MibHLH043)that may be involved in the synthesis of mango flavonoids were initially screened.And MibHLH054);3 genes(MibHLH035,MibHLH038 and MibHLH076)that may be involved in the regulation of mango pulp sugar accumulation;3 bHLH genes(MibHLH048,MibHLH078 and MibHLH090)related to low temperature stress;and 1 may be involved in postharvest fungal diseases The defensive gene MibHLH093.4.Genomic mapping analysis found that the mango WD40 s gene was unevenly distributed on 20 chromosomes.Phylogenetic analysis shows that it can be divided into 7subgroups.The same subgroup has similar conserved motifs,conserved domain positions and gene structure,but for the whole family,the sequence is less conserved.Collinearity and selective pressure analysis revealed that there are 48 pairs of collinearity relationships among the mango WD40 members.It is speculated that the expansion of the mango WD40 gene family may be closely related to the large chromosome replication event.The selection pressure analysis found that the Ka/Ks value of most collinearity genes was far less than 1,indicating that the mango WD40 gene may be mainly subjected to purification and selection during the evolution process.Expression analysis found that the expression of mango WD40 s in different tissues has a preference,but it was not found to be significantly related to genes related to anthocyanin synthesis.5.Genomic mapping revealed that Mi RAVs are distributed on 5 different chromosomes.Combined with Arabidopsis thaliana,tomato(Solanum lycopersicum),pepper(Capsicum annuum),sweet orange(Citrus sinensis)RAVs protein for phylogenetic analysis,they were divided into 5 subgroups.Mi RAVs proteins are distributed in subgroups II and IV,and there are similar conserved motifs and conserved domains in the same subgroup.Collinearity analysis revealed that Mi RAVs had 6 pairs of fragment duplication genes.It is speculated that the expansion of Mi RAVs family may be related to fragment replication events.Selection pressure analysis found that the value of Ka/Ks for collinearity genes was much less than 1,indicating that Mi RAVs may be mainly affected by purification and selection during the evolution process.The analysis of cis-acting elements showed that Mi RAVs contained a large number of light-responsive elements,followed by hormone-responsive elements,biotic and abiotic stress and growth and development-related elements.Expression analysis revealed that the expression of Mi RAVs in different tissues of the’Alphonso’ mango has a preference.Mi RAV2 and Mi RAV3 mainly play a role in the pericarp and may be involved in the regulation of pericarp anthocyanin synthesis.Mi RAV4 plays a role in the main fruit pulp.Mi RAV2,Mi RAV3 and Mi RAV4 are related to mango fruit development and maturation and response to adversity stress.Mi RAV3 is also related to ’Kent’ mango’s response to low temperature damage and stress and’Ataulfo’ mango fruit softening after hot water treatment.
