Rubber trees as a kind of important economic crop,latex is produced by one of the important source of natural rubber production.Natural rubber with synthetic rubber is no substitute for the physical and chemical properties,and therefore is widely used in many important fields.Rubber anthracnose is caused by Colletotrichum gloeosporioides,which is one of the main reasons affecting the decline of latex yield.The host range of C.gloeosporioides is very wide,and it has a variety of infection strategies for different plants.Therefore,it is difficult to control the pathogen.Transcription factors are the key factors in the regulation of gene expression.Through studying the functions of transcription factors,we can further analyze their role in the growth,development and pathogenic process of C.gloeosporioides.It provides an important theoretical basis for finding new fungicide targets of C.gloeosporioides and designing new control strategies for rubber anthracnose.In this study,one Cys2His2 and one Zn2Cys6 transcription factor were identified from C.gloeosporioides,named Cg Con7 and Cg Asw A,respectively,and analyzed by bioinformatics and phenotype analysis to clarify their biological functions.The research results are as follows:Bioinformatics analysis results show that the length of the ORF of Cgcon7 is 1335 bp and encoded a 444-amino acids protein,containing a C2H2 zinc finger domain.Compared with wild-type strains,the knock-out mutant showed slow vegetative growth,low conidial yield,significantly reduced spore germination rate,no appressorium,sensitivity to stress factors and reduced pathogenicity.The results showed that Cg Con7 is involved in the regulation of vegetative growth,conidiation,germination,appressorium formation and invasion,oxidative stress,osmotic pressure response and pathogenicity in C.gloeosporioides.Bioinformatics analysis results show that the length of the ORF of Cgasw A is 2142 bp and encoded a 713-amino acids protein,containing a GAL4 domain,an AT_hook domain and a fungal-specific transcription factor domain.Compared with the wild-type strain,the knock-out mutant showed slow vegetative growth,low conidial yield,delayed spore germination and appressorium formation,tolerance to SDS and weakened pathogenicity.The results showed that the Cg Asw A is involved in the regulation of vegetative growth,conidiation,germination,appressorium formation and invasion,cell wall integrity and pathogenicity in C.gloeosporioides.
