Background & objective: Copper is one of the trace elements necessary for the body composition.Intake of the appropriate amount of copper can promote the growth of the body and improve the immunity of the body.At the same time,the heavy metal copper is also highly toxic.Excessive intake of copper can cause damage to the body.Yinzhihuang granule is one of the commonly used clinical hepatoprotective drugs.In order to study the poisoning mechanism of chronic copper poisoning and the relieving and therapeutic effects of Yinzhihuang Granule on copper sulfate-induced liver injury in mice,the following studies were carried out.Method: This experiment takes daily feeding of drinking water containing copper sulfate as an example to simulate the liver damage model of mice caused by excessive copper in the natural environment,so as to explore the toxic effect of copper ions on the body and its mechanism of action.According to the modeling results,a suitable drug was selected to explore the therapeutic effect of the drug on liver injury in mice with copper poisoning.The study used methods such as monitoring the body weight and survival curve of mice during the modeling period,detecting blood biochemical indicators,observing histopathological sections,and transcriptome sequencing to explore the effects of copper poisoning on the body of mice.Then I chose Yinzhihuang Granule to study its therapeutic effect on liver damage in mice with copper poisoning,and its alleviating effect on liver damage in mice caused by copper poisoning.Results: The results of this experiment show that copper poisoning can inhibit the growth and development of mice,reduce survival rates,increase the levels of enzymes such as AST and ALT in the serum,and cause obvious pathological phenomena in the liver,such as inflammatory cell infiltration and hepatocyte degeneration;copper poisoning can also significantly up-regulate or down-regulate genes related to the liver,among which up-regulation Sult3a1,Cyp2d10,Hsd3b7,Maz,and down-regulated Gnas,Alas1,Elovl3,Meg3,Pbld2,Phyhd1,which proved that copper poisoning damages the liver of mice.Yinzhihuang Granule can delay the death of copper-poisoned mice for about 1 to 8 days.In the study of Yinzhihuang Granule in the treatment of liver injury in copper-poisoned mice,there was no death in the Yinzhihuang Granule administration group.Yinzhihuang granule can alleviate the weight loss trend of copper-poisoned mice,reduce the content of serum enzymes such as AST,ALT,and CK in the serum of copper-poisoned mice,improve the pathological symptoms of liver tissue,and accelerate the excretion of excess copper in the body.So as to alleviate liver damage in mice,it has a certain alleviation and treatment effect on copper poisoned mice.Conclusions: Copper poisoning can cause liver damage in mice.This experimental study found that the300 μg/mL and 1000 μg/mL test groups had the most severe liver damage.Yinzhihuang Granule were used to treat mice with liver injury caused by copper poisoning,and it was found that Yinzhihuang Granule had a significant alleviating and therapeutic effect on the liver injury of copper poisoning mice.
