| The potato(Solanum tuberosum L.)is the fourth largest food crop and is widely grown around the world.Potato is vμLnerable to virus infection,resμLting in the potato yield and quality decline.So we investigated the occurrence of potato viruses in potato-growing areas of Shanxi Province in order to better understand the characteristics of these potato viruses and to reduce their harm to potato yield and quality to a greater extent.The samples were tested by bio-chip,and we found that there were 5 viruses and 1 viroid circμLating in potato growing areas in Shanxi Province,namely PVY,PVS,PVM,PVX,PLRV and PSTVd.At the same time,Immuno-RT-PCR was used to identify PVY strains: seven strain types were detected,namely types of SYR-I,SYR-Ⅱ,SYR-Ⅲ,NWi-B,NWi-C,NTN-A and NTN-B.The main resμLts of the study are as follows:1.Using bio-chip and ELISA method,in north,middle and South of Shanxi province,a total of 319 samples with typical symptoms of potato virus were collected and tested,the infection of 5 viruses and a viroid were identified,based on the resμLts,PVY was able to detected from each region,and the highest of the occurrence of was PVY(87.77%),140 samples were detected as PLRV,with detection rate of 43.89%,114 samples were detected as PVM,with detection rate of 35.74 %,70 samples were detected as PVS,with detection rate of 21.94%,6 samples were detected as PSTVd,with detection rate of 1.88%,1 samples were detected as PVX,with detection rate of 0.31%,and 0 sample were detected as PVA.Among Huairen,Lanxian,Zuoyun,Kelan,Wenshui,Zuoquan,Puxian and Qinyuan,Lanxian had the lowest rate of PVY,the detection rate was 77.78%,Wenshui had the highest rate of PVY,the detection rate is 97.50%.Among the three regions of north,middle and South of Shanxi province,the PVY incidence was the highest in middle(94.29%),and the PVY incidence was the lowest in north(83.89%).In north area,the PVY incidence was the lowest rate in Lanxian(77.78%),and PVY incidence was the highest rate in Zuoyun(91.11%).In middle area,the PVY incidence was more than 90.00%.In south area,the PVY incidence was above 86.00%.Using Immuno-RT-PCR method,280 positive samples were typing as different strains,and the resμLts showed that 242 samples were infected by single strain,the detection rate was 86.43%,38 samples were infected by mixed strains,the detection rate was 13.57%.A total of 7 strains with different recombinant structures were identified,namely types of SYR-I,SYR-Ⅱ,SYR-Ⅲ,NWi-B,NWi-C,NTN-A and NTN-B.SYR-Ⅱ was the predominant strain in our province(36.43%),SYR-Ⅲ had the lowest detection rate(6.43%).In north,a total of 7 strains were detected,SYR-Ⅱ had the highest detection rate(15.89%),SYR-Ⅲ had the lowest rate(7.95%);In middle,a total of 5 strains were detected,SYR-Ⅱ had the highest rate(50.00%),NTN-B had lowest rate(3.03%);In south,a total of 5 strains were detected,SYR-Ⅱ had the highest rate(71.43%),SYR-I had lowest rate(1.59%).Among them,in Huairen,NWI-B was the predominant strain,the detection rate was 22.50%,in Lanxian,NTN-A was the predominant strain,the detection rate was 25.71%,in Zuoyun,the predominant strain was SYR-Ⅱ,the detection rate was 26.83%,in Kelan,NTN-B was the predominant strain,the detection rate was 31.43%,in Qinyuan,SYR-Ⅱ was the predominant strain,the detection rate was 62.16%,in Zuoquan,NWI-B was the dominant strain,the detection rate was 29.63%,in Wenshui,SYR-Ⅱ was the dominant strain,the detection rate was 74.36 %,and in Puxian,the SYR-Ⅱ was the dominant strain,the detection rate was 84.62%.2.Seven representative strains were sequenced and phylogenetic tree and recombination analysis were conducted,the data shown that: PVY-SX-HR-2 was 9686 nt in length and encoded 3061 amino acids and had 3 recombination breaking points;PVY-SX-WS-24 was 9681 nt in length,encoded with 3061 amino acids and had 4 recombinant breaking points;PVY-SX-QY-4 was 9681 nt in length,encoded 3061 amino acids and had 3 recombinant breaking points;PVY-SX-PX-24 was 9680 nt in length,encoded 3061 amino acids and had 4 recombinant breaking points;PVY-SX-KL-12 was 9676 nt in length,encoded 3061 amino acids and had 4 recombinant breaking points;PVY-SX-ZY-40 was 9680 nt in length,encoded 3061 amino acids and had 2 recombinant breaking points;The PVY-SX-LX-2 was 9683 nt in length,encoded with 3062 amino acids and had 2 recombinant breaking points.Combined with recombination and phylogenetic analysis,we found that: PVY-SX-ZY-40 belonged to the NWI-B,PVY-SX-LX-2 belonged to the NWI-C,PVY-SX-WS-24 belonged to the SYR-Ⅱ,PVY-SX-QY-4 belonged to the SYR-I,PVY-SX-HR-2 belonged to the NTN-A,PVY-SX-KL-12 belonged to the SYR-Ⅲ.PVY-SX-WS-24 belonged to the NTN-B.3.Using the predominant SYR-Ⅱ and NWi-B strains as inocμLum,pathogenic experiments were carried out on potato varieties Ke Xin 13,He 15,Ji Zhang 12,Jin 16,Qing 10 and Xi Sen 6.It was found that the Ke Xin 13,He 15,Ji Zhang 12 were successfμLly infected,the non-inocμLated systemic leaves showed different degrees of mosaic symptoms.Jin 16,Qing 10 and Xi Sen 6 were not infected. |
