| Being one of the most important economic species of China,since the success of total artificial reproduction of little yellow croaker(Larimichthyas polyactis)for the first time in2015,the breeding technologies have achieved great progress.While the current embryo hatching rate is still lower than 60%,which limits the further development of the small yellow croaker farming industry.The effects of spectra imposing on fish embryo development are species specific,to determine the influences of spectra on embryo development of little yellow croaker and its potential application value,the current study performed and finished the comparing analysis of the hatching rate,incubation speed,immunol level,rhythm regulation and proteomic analysis of little yellow croaker embryos treated under six different spectra: red(660-670nm)、 blue(455-465nm)、 green(515-525nm)、dark、full(400-780nm)and natural light.The results showed and:The hatching rate varied significantly between the groups,among which the red group had the highest hatching rate(49.56%),followed by the dark group(38.66%),the full group(31.46%),the natural group(31.25%),the green group(30.72%)and the blue group(29.58%).The embryos of each group showed significant differences in development speed since 13 hours post fertilization.The red and dark groups hatched most fast,followed by the natural group,the green group,the full group and the blue group.The expression levels of melanopsin and clock also varied significantly among groups,the former expressed highest in the blue group and lowest in the red group,which was opposite to clock.The concentration of Ig M was higher when light existing and highest in the red group.The complement C3 expressed higher in the natural,green and full light groups than in other groups.Both of TGF-β and IL-1β had the highest levels in the natural group,the former had higher levels in the dark and full groups than in the green,blue,and red groups,and the latter was the opposite.Taken the natural group as control,the proteomics of the embryos from the other 5spectral groups were analyzed.The GO and KEGG enrichment analysis showed,the major differences mainly focused on the t RNA metabolic process,like t RNA thio-modification,and t RNA-binding;In addition,the biosynthesis process of cellular nitrogen compounds,and sulfur relay systems were also the main differences.When it comes to each group,the blue group mainly enriched in sulfides and ubiquitin proteases,the dark group in ribosoproteins,the full group in ribosoproteins and peptidases,the red group in DNA topoisomerase,and the green group in epoxide hydrolases,where all occurred in nucleus.Based on the above experiments,the spectra imposed significant differences on the embryo incubation,immunity level,rhythm regulation of little yellow croaker,and the red light could promote embryo hatching,increase the transcription and translation level of immune factors such as Ig M,enhanced the expression of rhythm genes,and improve the level of transcriptase and energy metabolism enzymes of embryos. |
