Ziziphus jujuba Mill.is a plant of the genus Ziziphus Mill.in the family Rhamnaceae.It is an important woody grain tree species in China.and economic value.In production,wild jujube is often used as rootstock to improve the drought resistance,salinity resistance,barren resistance and disease resistance of jujube trees,so as to achieve the purpose of early fruiting,high quality and large yield.At present,there are few researches on jujube rootstocks.The existing researches focus on the cultivation of rootstocks and the improvement and improvement of grafting technology,while the root growth and development of jujube rootstocks,the compatibility of grafting,the internal relationship between the growth and development of rootstocks and their interaction the mechanism of cultivation is still unclear,which restricts the innovation of cultivation technology.In this paper,Ziziphus spina-christi and Ziziphus acidojujuba were used as test materials,and four grafting combinations were Zspina-christi/Z.acidojujuba(Scion/Rootstock,G/S),Z.acidojujuba/Z.acidojujuba(S/S),Z.acidojujuba/Z.spina-christi(S/G)and Z.spinachristi/Z.spina-christi(G/G),the grafting method is used for grafting,and the nongrafted Z.acidojujuba and Z.spina-christi are self-rooted seedlings(S,G)as a control,to study the effects of scion on root growth and morphological changes of rootstocks,as well as sugar content,enzyme activities related to sugar metabolism and other physiological levels,in order to provide a basis for the study of the interaction mechanism between the rootstock and ear of Jujube.The main findings are as follows:1.The scion of Z.spina-christi promotes the root morphological growth of the rootstock of Z.acidojujuba.(1)At 40-60 days after grafting,Z.spina-christi was used as a scion,and the lateral roots of the rootstock of Z.acidojujuba increased.(2)40-60 days after grafting,the total root length,total root surface area,root average diameter,root volume and root tip number of S.jujube as scion rootstock were 1.33%,16.89%,2.01%,1.33%,16.89%,2.01%,1.56%and 4.1%.(3)The total root length,surface area,average diameter,volume and number of root tips of the rootstock of jujube as a scion were significantly smaller than those of the root stock of Z.spina-christi during the growth process,indicating that the rootstock of jujube as a scion inhibited the growth of the root system of Z.spina-christi.2.The scion of Z.spina-christi had a significant influence on the anatomical structure of the main root of the rootstock of Z.acidojujuba.(1)Compared with the self-rooted seedlings of Zacidojujuba,the diameter of the main root and the thickness of the cortex of the rootstock increased by 11.2%and 5.8%when Zspina-christi was used as a scion.(2)After 60 days of grafting,the scion of Zspina-christi increased the diameter of the rootstock,the thickness of the cortex,the area of vascular column,the area of xylem and the area of phloem.The main root diameter and cortical thickness of G/S were 33.9%and 20.0%higher than those of S/S;the diameter and area of vascular column of G/S were 16.0%and 49.2%higher than those of S/S;The area of xylem and phloem of main root of G/S was 40.6%and 9.0%higher than that of S/S.(3)Z.spinachristi scion significantly increased the diameter of main root ducts and increased the number of ducts.The diameter of the main root ducts of the grafted seedlings G/S was 24.9%and 16.1%higher than those of the rooted seedlings grafted on this stock and Z.acidojujuba,and the number of main root ducts was 15.3%and 3.8%higher.(4)Zacidojujuba as a scion reduces the diameter and number of main root,thickness of cortex,diameter and area of vascular column,area of xylem and phloem,as well as diameter and number of vessels.3.The scion of Z.spina-christi increased the root activity,root sugar accumulation and related enzyme activities of the rootstock of Z.acidojujuba.(1)After 60 days of grafting,the root vigor of the grafted seedlings G/S and the self-rooted seedlings of Z.acidojujuba were increased by 14.9%and 25.2%,respectively.increased by 1.5%;the root vigor of the grafted seedling G/S rootstock was 1.12 times that of the rootstock grafted on this stock.(2)At 10 days after grafting,the root system of the grafted seedlings was affected by the scion,and the activities of sugar and enzymes were lower than those before the grafting;after 60 days of grafting,the content of sucrose and glucose in the G/S roots of the grafted seedlings was higher than that of the grafted stock 6.34%,14.07%;Z.spina-christi was used as a scion rootstock for sucrose phosphate synthase(SPS)activity,sucrose synthase(SuSy)activity,hexokinase(CWIN)activity and cell wall invertase(HXK)activity.The root system of grafted rootstock was 13.26%,6.13%,1.98%and 3.15%higher than that of this stock to varying degrees.
