Sugarcane is an important sugar crop.And its always faced with many challenges of biotic and abiotic stress,especially drought and cold stress which restrict sugarcane production.DNA methylation is a highly conserved epigenetic modification,which plays an crucial role in gene expression regulation.transposon silencing and genomic imprinting.DNA methylation is dynamically regulated by DNA methyltransferase.Cumulative evidence have shown that DNA methylation is not only involved into plant growth and development,but also plays an important role in plant response to abiotic stress.In recent decades,DNA methyltransferase gene family has been extensively studied in many plants,but there is no any studies on DNA methyltransferase gene family in sugarcane due to the complexity of sugarcane genome.Here,we combined with bioinformatics approach to characterize DNA methyltransferase genes and analyze the expression across different tissues and treatments from abiotic stress.We have obtained many progresses as below:1.28 DNA methyltransferase genes were identified by the combination between sequence similarity and conserved functional domain.Then we also performed subfamilies classification,domain identification,chromosome localization andgene duplication.2.According to the phylogenetic tree analysis of DNA methyltransferase gene family members in 16 different species,which means that DNA methyltransferase genes may have played an important role in the evolution of plants.At the same time,the genome duplication event and the remarkable amplification of DRM subfamily genes are the key to the amplification of DNA methyltransferase gene family in S.spontaneum.3.Combined with the transcriptome data of root,stem bark,stem core and leaf,three SsDNA methyltransferase genes(SsMET1b-1A,SsCMT3a-1P,SsCMT3a-2B)were highly expressed in root tissue,but were low expressed in other tissues,which indicated that these three sugarcane DNA methyltransferase genes might play an important regulatory role in root growth and development.However,the expression of DNA methyltransferase gene showed dynamic changes under drought and cold stress.Token together,we have made use of sugarcane genome sequence,combined with molecular evolution,transcriptomics and co-expression network analysis,to carry out a series of analysis on the identification and transcription regulation of sugarcane DNA methyltransferase gene.our results will shed lights into the dynamics of epigenetic modification across different tissues and abiotic stress treatments,and provide important resources for sugarcane breeding.
