Background: Canine Mammary Tumor(CMT)is the most common tumor in female dogs,which seriously threatens the health of dogs.The study of CMT will help to improve the survival rate of dogs.Surgical resection is the preferred treatment for canine mammary tumors,but the postoperative prognosis is poor.Adjuvant chemotherapy is required after surgery.However,the types of drugs used for canine mammary tumor chemotherapy are limited,and there are certain drug resistance phenomena.Abundant traditional Chinese medicine is an important resource for screening anti-tumor drugs.Celastrol(Cel)is an extract of the Chinese herbal medicine Tripterygium wilfordii,which has great application prospects for the treatment of tumors.At present,the effect and mechanism of celastrol on canine mammary tumor are not clear.Method: In this study,the inhibitory effect of celastrol on canine mammary tumor cells in vitro was examined by CCK8,cell colony,migration and invasion assays.Flow cytometry,q PCR and Western Blot were used to explore the anti-tumor cell mechanism of celastrol in vitro.Finally,by establishing a tumor-bearing mouse model,the anti-canine mammary tumor effect of celastrol in vivo was explore.Results:(1)Celastrol can inhibit the proliferation of canine mammary tumor cells in vitro,and reduce the migration and invasion ability of canine mammary tumor cells.(2)Celastrol promoted cell apoptosis by up-regulating the protein expressions of Cleaved Caspase-3 and Cleaved Caspase-9,and induced cell cycle arrest by regulating cell cyclerelated proteins.(3)Celastrol was highly toxic in vivo,and its anti-tumor effect was difficult to exert.Conclusion: Celastrol induces apoptosis by activating the Caspase signaling pathway,and induces cells to arrest in the G2/M phase,thereby exerting anti-canine mammary tumor cells.However,it did not significantly inhibit the tumorigenic ability of canine mammary tumor cells in vivo.This study revealed the effect and partial mechanism of celastrol on inhibiting canine mammary tumors in vitro,provided a new direction for anti-canine mammary tumor drugs,and provided an important theoretical reference for the clinical application of celastrol.
