| Morus species as an important economic crop,there have always been different views on classification systems.The chloroplast and mitochondrial genomes are important components of plants complete genome and are widely used to study the phylogenetic analysis of plants.In this study,the cultivated species of morus multicaulis Perr “Husang 32”and Morus atroppurpurea Roxb “Lunjiao 40” were sequenced by high-throughput sequencing technology to identify the chloroplast and mitochondrial genomes,combined with published genomic data in the Gen Bank database,to perform a comprehensive genomic and phylogenetic analysis.The results are as follows.1、The results of sequencing and analysis of the chloroplast genome of mulberry:The mulberry chloroplast genomes typical tetrads.The chloroplast genome of M.multicaulis “Husang 32”size is 158,817 bp with a CG content of 36.24%.In addition,the large single-copy region(LSC)was 87,880 bp,and the small single-copy region(SSC)was19,835 bp.Furthermore,the inverted repeat region(IRa/IRb)was 25,551 bp.In the M.atroppurpurea“Lunjiao40”,the chloroplast genome is 158,776 bp,with a GC content of36.26%.The SSC and LSC lengths are 19,750 bp and 87,670 bp,respectively.In addition,the IR region length is 25,667 bp.The two morus species produced 113 annotated genes,including 79 protein-coding genes,30 t RNAs and 4 r RNAs.The protein-coding genes were about 43% of the cp DNA.The genes and the CG content of the chloroplast genomes were similar to other morus plants.The phylogenetic tree and clustered analysis of the dicotyledonous plants and morus using the chloroplast genome sequence reveals that the morus were closely related to Rosaceae.The M.multicaulis and M.atroppurpurea clustered together,it’s means there is a closer relationship.Further study of the Psb A gene sequence containing SNP loci and the phylogenetic analysis in morus reveals that the results show morus is monophyletic,which provided a new idea for species identification and phylogenetic analysis of mulberry.2、The results of sequencing and analysis of the mitochondrial genome of mulberry:The mulberry mitochondrial genomes are circular.The length of M.multicaulis“Husang 32 is 361,546 bp,with the CG content being 45.42%.In addition,54 genes were annotated,including 31 protein-coding genes,3 r RNAs and 20 t RNAs.The M.atroppurpurea “Lunjiao40” mitochondrial genome is 395,412 bp in size,C G content of45.50%,and 57 genes,including 32 PCGs,22 t RNA and 2 r RNA.The coding region of the gene accounted for 8% of the mt genome,and the non-coded sequence accounted for the vast majority of the genome.Furthermore,212 scattered repeats were identified in M.multicaulis,accounting for 8.93%,and 216 scattered repeats in M.atroppurpurea accounted for 9.44% of the mt genome.The DNA migration from chloroplast to mitochondrial genomes accounted for 7.80% and 8.56% of the mt genome.Also,377 and 373 RNA editing sites were predicted in M.multicaulis and M.atroppurpurea.Finally,the phylogenetic analysis of the mt genome sequence of Morus and other families was clustered.The three species of morus get together and then get together in one group with the Leguminosae.In this study,high-throughput sequencing technology was used to sequence,assemble,and annotate mulberry’s chloroplast and mitochondrial genomes and conduct a comprehensive genomic analysis to provide a new idea for future studies on phylogenetic analysis and identification of mulberry,also enriches the genome-wide database of morus. |
