Fish maw(dried swim bladder),rich in collagen,is a traditional Chinese medicinal and edible tonic.Nibea coibor is an important economic fish that produces the precious fish maw "white fish maw".In recent years,the market for high-quality fish maw has become increasingly strong,with the improvement of people’s material living standards and in-depth understanding of the nutritional and health care value of fish maw.Collagen is the main nutrient component of fish maw and increasing the deposition of collagen in swim bladder helps to improve the yield and quality of fish maw,which has important practical significance.Studies have shown that Eucommia Ulmoides and faba bean can promote the collagen deposition in muscles such as grass carp.However,it is still unclear whether Eucommia Ulmoides,faba bean and their extracts can promote the collagen deposition in the swim bladder of Nibea coibor.In the study,Eucommia Ulmoides leaf powder(2% and 4%)and Eucommia chlorogenic acid(200mg/kg and400mg/kg),faba bean alcohol extracts(0.9%,1.8% and 3.6%)and faba bean water(1.25%,2.5% and 5%)extracts have been added to the feed to carry out 10-week breeding experiment to figure out the effects on the growth performance,antioxidant capacity and the deposition of collagen in the swim bladder which would provide scientific basis for the development of compound feed additives to improve the yield and quality of fish maw in Nibea coibor.The main research contents and results are as follows:1.Effects of Eucommia Ulmoides leaf powder and Eucommia chlorogenic acid on growth performance,antioxidant capacity and collagen deposition of swim bladderIn terms of growth performance,,the addition of 2% Eucommia Ulmoides leaf powder to the feed significantly reduced the weight gain rate and specific growth rate of fish,and increased the feed coefficient,while the addition of 4% Eucommia Ulmoides powder had no significant effect on the above growth performance,but significantly increased the swim bladder weight;the feed added 200 mg/kg Eucommia Ulmoides Chlorogenic acid significantly increased fish body weight gain,specific growth rate,swim bladder weight and decreased feed coefficient,while 400mg/kg Eucommia chlorogenic acid group had no significant effect on growth performance.In terms of antioxidant capacity,adding 2% and 4% Eucommia Ulmoides leaf powder to the feed significantly increased serum T-AOC,GSH-PX and liver SOD activity,and decreased the MDA content in the liver;adding 2% Eucommia Ulmoides leaf powder to the feed significantly increased serum MDA and CAT activities,and 4% The trend of Eucommia Ulmoides leaf powder is opposite;adding 200mg/kg Eucommia chlorogenic acid to the feed can significantly reduce the content of MDA in serum and liver,and increase the activities of serum GSH-PX and CAT;adding 400mg/kg Eucommia chlorogenic acid can significantly reduce MDA content in serum and liver and improve the activities of serum T-AOC,GSH-PX,CAT and SOD and CAT in the liver.In terms of intestinal digestive enzymes,adding 2% and 4% Eucommia leaf powder and 200mg/kg and 400mg/kg Eucommia chlorogenic acid to the feed significantly increased the lipase activity and decreased the amylase activity.For terms of collagen deposition,only 400mg/kg Eucommia chlorogenic acid group significantly increased the expression levels of col1a1 and col1a2 genes and collagen content in swim bladder,which promoted collagen deposition in swim bladder.2.Effects of alcohol and water extracts of faba bean on growth performance,antioxidant capacity and collagen deposition of swim bladderIn terms of growth performance,the addition of faba bean alcohol extract(1.8%,3.6%)to the feed significantly reduced the fish body weight gain rate,specific growth rate and swim bladder weight,and increased the feed coefficient.The addition of faba bean water extract(1.25%,2.5% and 5%)to the feed significantly increased the weight gain rate,specific growth rate and swim bladder weight,and decreased the feed coefficient.In terms of antioxidant capacity,the addition of 0.9% and 3.6% faba bean alcohol extract to the feed significantly reduced the serum MDA content and SOD activity.In the 0.9% faba bean alcohol extract group,the activities of serum T-AOC and CAT were significantly increased.In addition,0.9% and 3.6% of faba bean alcohol extract significantly increased liver MDA content,SOD and CAT activities.The addition of faba bean water extract(1.25%,2.5% and 5%)to the feed significantly reduced the serum MDA content and SOD activity.In addition,the addition of 1.25%faba bean water extract significantly increased the activities of serum T-AOC,GSHPX,CAT and SOD and CAT in liver.In terms of intestinal digestive enzymes,the addition of faba bean alcohol extract(0.9%,1.8%,3.6%)and faba bean water extract(1.25%,2.5%,5%)to the feed significantly increased the lipase activity and decreased the amylase activity.In terms of collagen deposition,the 1.25% faba bean water extract group significantly increased the expression level of col1a2 gene and collagen content,which promoted collagen deposition in the swim bladder.
