| This study took Xinjiang grape virus disease as the research object.Through field survey statistics and metagenomic sequencing combined with RT-PCR,the occurrence characteristics,epidemic trend and damage degree of grape virus disease were understood,and the types of grape virus in Xinjiang and the situation of compound infection were identified.It lays the theoretical foundation for the effective prevention and treatment of virus disease.The main findings are as follows:1.Virus disease in xinjiang grape growing region of field investigation,the main performance etiolation,spots,the symptom such as green,deformity,according to the different symptoms of characteristic,multiplicity of symptoms are complex symptoms,in order to specify the type of grape virus and virus,this study deals with the method of macro genome sequencing of xinjiang grape virus diseases of pathogen identification of samples,Through Taxonomic analysis contigs and open reading frames(ORFs),annotated viruses and extracted virus sequences to determine the species of Grape viruses in Xinjiang.A total of 7 RNA viruses were detected.GLRa V3,GINV,GFab V,GRSPa V,GVA,GPGV and GFKV respectively.2.Grape viruses and viroids could be detected in 7 grape growing areas in Xinjiang.Six RNA viruses GFab V,GINV,GLRa V3,GFKV,GPGV and GVA,one DNA virus GGVA and four viruses GYSVd1,GYSVd2,HSVd and AGVd were identified through molecular identification.GGVA,GINV,and GFab V are detected for the first time.Blast alignment sequences and evolutionary analysis were used to determine the possible genetic relationships of corresponding viruses.3.Virus detection was carried out on 304 suspected virus infection samples collected from 7 grape growing regions.The detection rate was different in different regions of Xinjiang.There were 64 strains without virus,and the virulence rate of plants was 78.95%.The detection rates of GFab V,GLRa V3,GINV,GPGV,GFKV,GVA and GGVA were 1.32~39.47%.The detection rates of AGVd,GYSVd2,HSVd and GYSVd1 viruses ranged from 18.75 to 55.26%.4.Compound infection of grape virus diseases was common in seven grape growing regions,including three conditions:(1)There are 24 combinations of the two viruses in the composite infection,among which the GINV+GFKV combination has the highest detection rate of 32.64%;the GGVA+GFab V combination has the rock-bottom detection rate,was 1.49%;there were 10 combinations of three or more virus combined infection,among which GPGV+GFKV+GVA combination had the supreme positive rate of 15.45%;GLRa V3+GINV+GFab V+GPGV+GFKV combination detected The lowest rate was 1.49%(2)There are 5combinations of two viroids combined infection,among which GYSVd1+HSVd combination has the topmost detection rate of 72.50%;GYSVd2+AGVd combination has the minimum detection rate of 24.36%;three or more viroids There were 5 combinations of compound infection,among which GYSVd1 +GYSVd2+HSVd combination had the highest detection rate of 68.75%;GYSVd1+GYSVd2+HSVd +AGVd portfolio had the minimum detection rate of 5.99%.(3)There are 27 combinations of A virus and one viroid combined infection,among which GFKV+HSVd combination have the supreme detection rate of 51.28%;GPGV+GYSVd2 combination has the lower-most detection rate of 2.56%;There were 41 combinations of multiple viruses and multiple viroids,among which GINV+GYSVd1+GYSVd2 and GINV+GYSVd1 +GYSVd2+HSVd+AGVd have peak detection rates,both 40.00%;The combined detection rate of GINV+GPGV+GFKV+GVA+GYSVd1+GYSVd2+HSVd+AGVd was the bottommost and minimum at 1.39%. |
