| Objective:With the increasing contradiction between the scarcity of forage and the high quality development of the farming industry,the full and rational use of agricultural waste such as crop straw is an effective way to solve the shortage of roughage.The best way to preserve nutrients and improve the efficiency of livestock use is to ferment grape leaves.In view of this,this study was conducted to investigate the species,physiological and biochemical characteristics,acid production rate and growth rate of the main naturally attached lactic acid bacteria in grape leaf silage,to select the best performing strains of lactic acid bacteria and molasses to add to grape leaf silage,to select the best combination of grape leaf silage in terms of nutritional quality,fermentation quality and silage processing,and to prepare grape leaf microsilage using the wrapped silage processing mode to replace some of the whole plant silage.The effect on growth performance,slaughter performance,serum antioxidant index and economic efficiency of fattened sheep was investigated to provide a scientific basis for the actual production of fattened sheep using grape leaf silage as a feed resource.Methods:By using traditional microbial culture methods and 16 S r DNA sequence analysis to identify microorganisms naturally attached to grape branches and leaves,four excellent lactic acid bacteria Q1,Q2,Q4 and Q5 were screened according to physiological and biochemical characteristics,acid production rate and growth rate.Combined with molasses,it was added to the silage of grape branches and leaves,and samples were taken on the 3rd,7th,14 th,30th,and 60 th days of the silage process to determine the nutritional quality and fermentation characteristics of hybrid paper mulberry silage,and to obtain the optimal silage mode for wrapping silage processing,Part of the roughage in the mixed daily diet of fattening sheep mainly based on whole-plant silage corn was replaced,and its effects on the growth performance,slaughter performance,serum antioxidant indexes and economic benefits of fattening sheep were explored.Results:(1)5 strains of lactic acid bacteria were obtained,of which the Q1 strain was identified as Leuconostoc mesenterica,the Q2 and Q5 strains were identified as Enterococcus faecium,and the Q3 strain was identified as Enterococcus casseliflavus,the Q4 strain was identified as Enterococcus hirae.The selected lactic acid bacteria can grow at p H 3~9,4~45℃,3% and 6.5% Na Cl concentration except Q1 strain which cannot grow at p H 9;among which Q4 strain is cultivated to 24℃.After h,the p H of the bacterial solution could reach 4.1,and the OD value of the solution was 1.4,showing the strongest growth and acid production ability.(2)By adding lactic acid bacteria and fermentation substrate molasses to the fermented feed,with the prolongation of fermentation time,the contents of DM,CP,WSC,NDF,and ADF in the L+M group were significantly higher than those in the control group,that is,other treatment groups.The change of,that is,the treatment of L+M group has the best effect,which significantly reduces the content of NDF and ADF,and significantly increases the content of DM,CP and WSC.Compared with the other treatment groups and the control group,the treatment group not only increased the content of LA but also decreased the content of AA and PA,so it was selected as the optimal silage exogenous addition treatment.(3)The treatment group was used as a mixed diet experimental group that replaced part of the forage in the control group with wrapped grape branches and leaves,and there was no significant difference in growth performance,slaughter performance,and serum biochemical indicators from the control group.The main fattening sheep mixed with roughage in the daily diet did not affect the growth performance,slaughter performance and serum biochemical indicators of the fattening sheep.Compared with the control group,the fattening profit of the PT group decreased by 15.71 yuan per animal,indicating that the replacement of part of the forage in the mixed daily diet of fattening sheep mainly based on silage corn will improve the economic benefits of fattening sheep breeding.Conclusion:Through the screening of naturally attached lactic acid bacteria on grape branches and leaves,it is mixed with molasses to form the best silage mode of grape branch and leaf silage fermentation quality and nutritional quality,and silage is made by using the wrapping silage technology to replace part of the fattening corn-based silage.The roughage in the sheep mixed daily feed will improve the economic benefits of fattening sheep breeding,that is,increase the fattening profit. |
