Phyllocnistis citrella Stainton(Lepidoptera: Phyllocnistidae),is an important pest that seriously harms citrus production and is widely distributed in major citrus producing areas in China.In recent years,the occurrence of leaf miner pests has become more and more serious,and how to effectively control them is an urgent practical problem to be resolved.In this thesis,the volatile components of ponkan flowers and leaves were identified by GC-MS,and the potential components were screened.Electroantennography(EAG)and field preference trapping experiments were carried out on the female and male adults of P.citrella.In addition,the dissected female and male adults of P.citrella were collected,and the transcriptome data was analyzed by Illumina high-throughput sequencing to screen the genes related to olfaction,which provided a theoretical basis for exploring the olfactory molecular recognition mechanism of P.citrella.The main findings are as follows:1.The GC-MS results showed that 86 and 109 volatile substances were identified from the flowers and leaves of Ponkan,covering compounds such as esters,ethers,phenols,aldehydes,alcohols,terpenes,olefins,and aromatic hydrocarbons.The highest content were terpenoids.The common compounds of flower volatiles and leaf volatiles were monoterpenes,terpenes and sesquiterpenes,and there were 17 kinds of compounds in total,including thujorene,β-pinene,basilene,linalool,terpinene oleene,alloocimene,citronellal,indolizine,geraniol,citral,methyl anthranilate,β-elemene,γ-elemene and Δ-juniperene,etc.;The content of volatile components was also different between them,Linalool content was the highest in flowers,up to 17.38% and β-hydroparnene content was the highest in leaves,up to 13.94%,but the main components of the top ten volatile compounds were aromatic compounds or aromatic compounds.2.According to the measurement results of GC-MS,five plant volatiles,basilene,linalool,(±)-limonene,myrcene and phenethyl alcohol were selected to test the EAG responses of P.citrella.The test results showed that,at a dose of 1μL,five plant volatiles could produce potential responses to the stimulation of the antennae of the female and male adults,among which the alcohol EAG was the highest,and the terpene alcohol EAG Second,terpenes had the lowest EAG.The stimulation potential of phenethyl alcohol to male antennae was significantly higher than that of basilene and(±)-limonene,and myrcene was the lowest to female antenna stimulation potential,which was significantly lower than that of phenethyl alcohol and linalool,and the sensitivity of the antennae to myrcene was significantly different between the sexes,the antennal potential response of males was 1.25 times that of females.3.The results of field trapping experiments showed that five single plant volatile attractants had poor field trapping effect on P.citrella adults,and basically had no attracting effect at two test doses(0.1μL and 1μL).Plant volatiles and sex pheromone synergistically attract adults with the increase of dose,and the effect of sex pheromone combined with plant volatiles and sex pheromone alone in attracting males was significant compared with the blank control difference,and the number of males co-baited is 45-70.5 times(0.1μL dose)and 66-108 times(1μL dose)of the blank control;the number of males attracted by pheromone alone is 42.5-69.5 times that of the blank control.Compared with the single sex pheromone lure,some plant volatiles such as basilene and linalool synergised with sex pheromone to attract more males.The numbers of males attracted by the basilene synergistic lure were 1.64 times(0.1μL dose)and 1.55 times(1μL dose)than that of single sex pheromone lure,the numbers of males attracted by the linalool synergistic lure were 1.66 times(0.1μL dose)and 1.53times(1μL dose)than that of the single sex pheromone lure,and the bait amount of the two groups of treatments was significantly higher than that of the other combinations.4.A total of 18 419 genes were successfully annotated through the comparative analysis of the transcriptome data of the whole worm and the decapitated of the female and male adults of P.citrella,and 17 odorant-binding protein OBP genes were obtained from annotation analysis,3 chemosensory protein CSP genes,45 odorant receptor OR genes,22 ionotropic receptor IR genes and 2 sensory neuron membrane protein SNMP genes.Pathway annotation of these genes revealed that they may be involved in metabolic pathways such as ko00561,ko00564,ko04080,and ko04141.Screening of olfactory-related genes from differentially expressed genes,it was found that 9 OBP genes,3 CSP genes,7 OR genes,4 IR genes and a SNMP gene were differentially expressed in the female and male adults.
