Taking Caenorhabditis elegans(C.elegans)as a biological model,the anti-aging activities of Orobanche cernua Loefling extract(OLE)and related molecular mechanisms were studied through the effects on related physiological indicators,mutant C.elegans,and related gene expression to provide data support and theoretical basis for the study of traditional Chinese medicine anti-aging preparations.The result showed that:1.N2 wild-type C.elegans tolerated a maximum concentration 7.813 mg/m L tolerated a maximum concentration of OLE.2.Compared with the control group,all dosing groups significantly prolonged the life span of Caenorhabditis elegans(P<0.05).Each dosing group had no significant effect on body length(P>0.05),Significant increase in pharyngeal pump frequency and body flexion on days 7 and 11(P<0.05),no significant effect on reproductive capacity(P>0.05);lipofuscin content was significantly reduced(P<0.05),significantly decreased reactive oxygen species(ROS)content(P<0.05),significantly increased catalase(CAT)and superoxide dismutase(SOD)content(P<0.05)and significantly decreased malondialdehyde(MDA)(P<0.05),significantly increased life span under oxidative stress in Caenorhabditis elegans(P<0.05),significantly increased life span under heat stress in Caenorhabditis elegans(P<0.05).3.The ratio of daf-16 to skn-1 entry in the cells of Caenorhabditis elegans was significantly increased after administration compared to the control group(P<0.05).In the mutant strain elegans lifespan assay,compared with the control,CB1370,CF1038,TJ1052,PS3551,QV225 mutant C.elegans did not significantly extend their lifespan after administration(P>0.05).DA1113,RB1206,CF1903,mutant C.elegans significantly extended their lifespan after administration(P<0.05);the expression of daf-2,age-1,akt-1were significantly reduced(P<0.05)and the expression of daf-16,mtl-1,sod-3,ctl-1,hsf-1,hsp16.2,skn-1,gcs-1 were significantly increased(P<0.05).4.Compared with the control group,neither CF1038 nor QV225 mutant C.elegans could significantly extend their lifespan after administration(P>0.05),and CB1370 and PS3551 had significantly higher survival rates after administration(P<0.05);the expression of daf-16,hsf-1 and skn-1 in N2 C.elegans was significantly higher after oxidative stress(P< 0.05),and the expression of daf-2 was significantly lower(P<0.05).Conclusions:1.OLE can improve anti-ageing related physiological indicators of Caenorhabditis elegans,improve antioxidant capacity and prolong lifespan.2.The mechanism by which OLE prolongs nematode lifespan is mainly through the insulin signaling pathway(IIS),but not dependent on the reproductive signaling pathway,diet restriction pathway(DR)and m TOR signaling pathway.3.OLE improves C.elegans resistance to oxidative stress,which is not entirely dependent on daf-2 and hsf-1,but mainly on daf-16 and skn-1.
