seed-used pumpkin(Cucurbita pepo L.),an annual vine herb of Cucurbitaceae,is a characteristic cash crop in Inner Mongolia.In recent years,due to the widespread occurrence of virus diseases,the yield and quality of seed-used pumpkin(Cucurbita pepo L.)have decreased seriously,which has become an important reason restricting the production of seed seed-used pumpkin(Cucurbita pepo L.).Taking leaf and seed of seed-used pumpkin(Cucurbita pepo L.)as materials,the pathogen of virus disease in Inner Mongolia was identified by RT-PCR,DNA sequencing and q PCR in order to clarify virus types of seed-used pumpkin(Cucurbita pepo L.).On this basis,carrying virus seeds were treated with physical methods and chemical agents to screen out effective seed detoxification methods.The research will provided a theoretical foundation for the prevention and control of seed-used pumpkin(Cucurbita pepo L.)virus disease in Inner Mongolia.The main results are as follows:1.By collecting the leaves with typical mosaic symptoms,it was detected that the main types of virus diseases from leaf in Inner Mongolia were CMV,ZYMV and WMV.2.Seeds of six varieties of seed-used pumpkin(Cucurbita pepo L.)with typical virus disease symptoms were detected by RT-PCR and the specific bands of CMV,ZYMV and WMV were amplified.The pathogens of seeds were identified as CMV,ZYMV and WMV in Inner Mongolia.3.Through the construction of phylogenetic tree,it was found that the CMV and WMV on leaves and seeds have the closest genetic relationship with the isolates from Japan and South Korea,respectively;ZYMV is closely related to the isolates of other crops in China.4.Seed were detoxified by five treatment methods including warm soup treatment,dry heat treatment,trisodium phosphate treatment,hydrochloric acid treatment and medicament(sunami)treatment.The results showed that five treatment methods could reduce the amount of virus in seeds in different degrees,and the detoxification rate of seeds was 62.80%-99.89%.
