| Ningxia Hui Autonomous Region is the main production area of Zhongwei goats. It has an irreplaceable ecological, social and economic status in the northwest region and has a high breeding value. It is the first batch of national livestock and poultry genetic resources protection species in China and has been included in the list of livestock and poultry genetic resources protection in Ningxia. It can be seen that the market potential of the Zhongwei goat is great and the breeding prospects are extremely optimistic. However, sheep infected with Chlamydia and its arboviruses can cause serious economic losses and hinder the development of livestock farming.Chlamydia(Chlamydia)is a class of infectious hosts, complex pathogenic characteristics of Gram-negative strictly intracellular parasitic microorganisms, widely distributed around the world, posing a serious threat to human and animal health. The Chlamydia epidemic is also more serious in China, and infection in sheep can cause typical symptoms such as abortion or stillbirth(weak fetus)in ewes, orchitis in males, and arthritis and encephalitis in newborn lambs.Chlamydia abortus is the main pathogen causing endemic abortion of ewes(EAE),which can cause widespread abortions in goats and other ruminants once the disease is prevalent.Arbovirus is a group of pathogenic microorganisms that are transmitted by arthropods to hosts such as vertebrates through their bites, mainly affecting ruminants, and its vectors are numerous and widely spread around the world. In this study,we investigated the epidemiology of chlamydia and its arboviral pathogens in the farming environment of Zhongwei goats, and provided a scientific and theoretical basis for the prevention and control of chlamydiosis and arboviral diseases in Zhongwei goats through the epidemiological investigation of chlamydia and the monitoring of arboviral pathogens in its farming environment. The specific findings are as follows:1.Serological survey of Chlamydia chlamydia in Zhongwei goatsIn this study, seroepidemiological investigation of Chlamydia was conducted on blood samples collected from the Zhongwei goat selection farm using enzyme-linked immunosorbent assay(ELISA).The results showed that the overall positive rate of Chlamydia serum antibodies in Zhongwei goats was 37.59%;the antibody positivity rate of Chlamydia in Liu Zhanjing breeding farm and Zhongwei core farm was 26.25%and 42.47%,respectively;the positive rate of Chlamydia in rams was 30.77%,and the positive rate of Chlamydia in ewes was 38.33%.2.Molecular epidemiological investigation of Chlamydia in Zhongwei goatsIn this study, nucleic acid tests for Chlamydiaceae and Chlamydia abortus were performed using fluorescence quantitative PCR on negative and rectal swabs collected from selected farms of Zhongwei goats. The results showed that the overall positive rate of Chlamydia section nucleic acid test in Zhongwei goats was 19.44%;Chlamydia abortus accounted for 4.76%of the positive samples of Chlamydia section;for the 108 individuals collected, the positive rate of Chlamydia abortus was 0.93%;the positive rate of Chlamydia section in rams was 14.71%and in female goats was 21.62%;the positive rate of Chlamydia abortus in rams was 0%and 1.35%positive for Chlamydia abortus in females.3.Detection of arboviruses carried by insects in the breeding environment of Zhongwei goatsIn this study, insects collected from the breeding environment of Zhongwei goats were tested for insect-borne pathogens using conventional PCR methods. The PCR assay and sequencing result analysis of the collected samples showed that in all samples,no insect vector diseases such as Chlamydia,foot-and-mouth disease, peste des petits ruminants, sheep infectious pustulosis, sheep infectious pleuropneumonia, anaplasma,Burkholderia spirochetes, rickettsia,Babesia protozoa, and Q fever rickettsia were detected. In conclusion,Chlamydia infection in Zhongwei goats is more serious, and there are obvious differences in infection in different fields and sexes, which require certain scientific control measures to be taken to control it.However, the presence of other pathogens cannot be ruled out and needs to be taken into account.Once some arboviral diseases occur, they spread rapidly and cause great losses. Therefore, the prevention and control of insect-borne diseases is a matter of urgency, and it is still necessary to master the biological characteristics of the pathogens of insectborne diseases, epidemiological trends, distribution and habits of vector arthropods,in order to take more effective measures to prevent and control the prevalence of insect-borne diseases,so as to better protect the excellent breed of Zhongwei goats. |
