| Pine sawdust wood vinegar is the liquid product obtained after condensation and separation of the vapor gas mixture derived from pine sawdust in the distillation equipment.It is rich in a variety of compounds and has the functions of antibacterial,antioxidant,regulating plant growth,anti-corrosion,etc.It is safe,pollution-free and low-cost,and is widely used in agriculture,forestry,medicine and health industry,food industry and many other fields.It has important practical significance.In this experiment,pine sawdust wood vinegar and mulberry leaves were taken as the research objects,the chemical composition and content of pine sawdust wood vinegar and mulberry leaf extract and their mixture treated with different organic solvents were analyzed,their effects on plant seed germination and antioxidant activities were studied,and wood vinegar with strong antioxidant activities was screened,and the synergistic antioxidant ability of combining with mulberry leaf ethanol extract was studied.The main findings are as follows:The effects of wood vinegar and mulberry leaves and their mixture on seed germination of radish,leek,Chinese cabbage and mustard were investigated.Overall,high concentration treatment had significant allelopathic inhibition on seed germination and seedling growth of 4plants.Ethyl acetate aqueous phase of wood vinegar(PWV3),hexane aqueous phase of wood vinegar(PWV4),the mixture of wood vinegar and mulberry leaves(MPWV)showed significant allelopathic effects on seed germination and seedling growth of the 4 plants.Under2000-fold dilution,both PWV3 and MPWV showed allelopathic effects on the germination rate of radish seeds,which were increased by 20.00%and 19.15%,respectively,compared with distilled water control group.When PWV4 was diluted 200 times,the germination rate of radish seed increased by 27.53%compared with the control group,the difference was significant.PWV3,PWV4 and MPWV showed allelopathic effects on stem length of leek seedlings under 500-fold dilution,which were increased by 30.60%,24.72%and 21.71%compared with control group,respectively.When the dilution ratio of PWV3,PWV4 and MPWV was 2000,the growth of stem length of Chinese cabbage seedlings was promoted by19.50%,13.24%and 12.51%,respectively,compared with the control group.At the same time,PWV3 and MPWV under 1000-fold dilution treatment showed allelopathic promoting effect on stem length of mustard seedlings,which was increased by 46.70%and 33.81%,respectively,compared with the control group,and the difference was statistically significant.The effects of wood vinegar,mulberry leaves,and their mixtures on the seed germination of radish,leek,pakchoi,and mustard were investigated.On the whole,under high concentration treatment,there was significant allelopathic inhibition on the seed germination and seedling growth of the four plants.Under low concentration treatment,wood vinegar in ethyl acetate aqueous phase(PWV3),wood vinegar in n-hexane aqueous phase(PWV4)The mixture of wood vinegar and mulberry leaves(MPWV)showed significant allelopathic effects on seed germination and seedling growth of the four plants.Under 2000 times dilution treatment,PWV3 and MPWV showed allelopathic promoting effects on the germination rate of radish seeds,which were significantly increased by 20.00%and 19.15%compared with the distilled water control group,respectively;However,the germination rate of radish seeds in PWV4 diluted at 200 times increased by 27.53%compared to the control group,with a significant difference.PWV3,PWV4,and MPWV showed allelopathic promoting effects on the stem length of leek seedlings under 500 times dilution treatment,which were 30.60%,24.72%,and 21.71%higher than the control group,respectively,with significant differences.When the dilution ratio of PWV3,PWV4,and MPWV was 2000,the growth of stem length of Chinese cabbage seedlings was allelopathically promoted,which increased by 19.50%,13.24%,and 12.51%compared with the control group,respectively,with significant differences.At the same time,PWV3 and MPWV under 1000 times dilution treatment showed allelopathic promoting effects on stem length of mustard seedlings,which increased by 46.70%and33.81%respectively compared with the control group,with significant differences and statistical significance.The polyphenol content and antioxidant index of three types of wood vinegar and ethanol extract from mulberry leaves were measured,and a strong antioxidant activity of wood vinegar was selected and mixed with ethanol extract from mulberry leaves in different proportions to explore the antioxidant activity of different groups and their antioxidant synergistic effects.The results showed that the n-hexane aqueous phase(PWV4)of pine sawdust wood vinegar had the strongest antioxidant activity among the three types of wood vinegar,with its scavenging DPPH·,·OH The IC50 value of ABTS+·and the EC50 value of total reducing power are the smallest among the three types of wood vinegar,respectively,32.085±0.0062 mg/m L,51.041±0.0132 mg/m L,18.474±0.0192 mg/m L,and 180.435±0.1564 mg/m L,respectively.The polyphenol content in wood vinegar(PWV4)is 49.92±0.13 mg/m L,which is the highest compared to the other two types of wood vinegar,with a significant difference.The antioxidant activity was the best when the composite ratio was PWV4:ME=1:8,and the IC50 values for scavenging DPPH·,·OH,ABTS+·and the EC50 values for total reducing power were the lowest among all composite ratio groups,respectively 0.198±0.0266 mg/m L,4.637±0.0083 mg/m L,1.195±0.0064 mg/m L,and 0.587±0.0064 mg/m L;In addition to scavenging·OH free radicals,all treatments at this complex ratio showed synergistic antioxidant effects. |
