| Gymnadenia conopsea R.Broyn is the dried tuber of the perennial herb Gymnadenia R.Br.in the Orchidaceae family,known as "Erikhtenai Gari" in Mongolian medicine and"Tibetan Wangla" in Tibetan medicine."It is mainly used in the treatment of deficiency of lung,kidney,qi and yin caused by asthma,lumbar and leg weakness.Studies have shown that the active ingredients of Panax ginseng have antioxidant,anti-allergic,anti-aging,memory improvement and protective effects on neuronal damage.In this study,hand palm ginseng polysaccharide was used as the study material and mouse peritoneal macrophages(RAW264.7)were used to investigate the effects of hand palm ginseng polysaccharide on macrophage toxicity/proliferation capacity,Fc/C3 b receptor function,phagocytosis,secretion function(NO,IL-6,IL-1β,TNF-α,IFN-γ,LZ);using hand palm ginseng polysaccharide as the study material and mouse peritoneal macrophages(RAW264.7)as the study target,Hoechst/PI staining combined with ELISA was used to investigate the effect of hand palm ginseng polysaccharide on LPS+ATP-induced system macrophage pyroptosis(Caspase-1,Caspase-3,IL-18,IL-1β).The results of the studies obtained were as follows.1.the mass concentration of 0.625 mg/mL~10 mg/mL of ginseng polysaccharide had the effect of promoting macrophage proliferation and phagocytosis,and the concentration of 10 mg/mL showed a maximum cellular value-added rate of 124.83% and phagocytosis rate of 113.11%.2.The mass concentration of 0.625 mg/mL~10 mg/mL of C.palmata polysaccharide inhibited the enhancement of phagocytosis of LPS macrophages and promoted the decrease of phagocytosis of HCSS macrophages,indicating a good bi-directional regulation of the function.3.The mass concentrations of 0.625 mg/mL~10 mg/mL of C.palmata polysaccharide promoted the Fc/C3 b receptor activity of macrophages(67.83% and 45% of YC and EA wreath rate,respectively),indicating that C.palmata polysaccharide has enhanced the immune adhesion function of macrophages.4.The mass concentrations of 0.625 mg/mL~10 mg/mL of ginseng polysaccharide showed a good positive effect on the secretion of NO,TNF-α,IL-1β,LZ and INF-γ by macrophages,and the reduction of the secretion function of activated macrophages showed a good bi-directional function.5.The polysaccharides of L.palmieri at mass concentrations of 0.625 mg/mL~10mg/mL showed different degrees of inhibition of LPS+ATP-induced macrophage scorching(inhibition of caspase-1,caspase-3,IL-1β,IL-18),with the best effect of inhibition of caspase-1 release at 2.5 mg/mL.The best effect of inhibiting the amount of Caspase-3,IL-1β and IL-18 release was observed at 5 mg/mL.The results of this study showed that the polysaccharide of Panax ginseng enhanced the immune adhesion and phagocytosis of macrophages by enhancing the activity of macrophage Fc/C3 b receptors,regulated the immune function of the body by promoting the secretion of macrophages,and enhanced the anti-inflammatory ability of the body by inhibiting the secretion of activated macrophages and the production of pyroptosis. |
