| Medicago falcata,also known as Wild Alfalfa,is a high-quality forage widely distributed in Northeast and Northwest China;Because Medicago falcata is a cross pollinated plant,it is easy to produce gene hybridization in the process of flowering and fruiting,resulting in the disappearance of some excellent characters.As a rapid propagation method of asexual propagation,cutting is of great significance for the optimal generation and breeding of Medicago falcata L In order to explore the factors affecting the cutting seedling rate of Medicago falcata,Medicago falcata was used as the research material in this experiment.The effects of cutting branch growth period,cutting branch leaf number,cutting matrix type,hormone concentration and matrix disinfection treatment on Medicago falcata cutting branches were set up respectively.By observing the growth rate of cutting branches,plant height,number of new leaves,root length and number of roots,the effects of different treatments on the cutting survival rate and growth of Medicago falcata were studied,so as to provide a reference basis for the asexual and rapid propagation of Medicago falcata.The test results are as follows:(1)The effect of growth period on cuttings: The fastest growth rate of plant height is the first flowering period;On the 15 th day,the number of new leaves was the most at the budding stage;In the 30 th,the number of new leaves was the most at the initial flowering stage.On the 15 th day,the bud stage had the largest number of roots;On the 30 th day,the branching stage had the largest number of roots.The highest survival rate was in the budding stage,which is 77.33%.(2)The influence of leaf number on cutting branches: The branches of Medicago falcata with leaves are higher than those without leaves,and the survival rate of Medicago falcata with leaves is the highest,which is 90.67%.(3)Effects of substrate types on cuttings: Flower soil has the fastest growth rate of plant height and the largest number of newborn leaves.On the 15 th day,the largest number of roots is concrete;On the 30 th day,the flower soil had the largest number of roots.The highest survival rate is flower soil,which is 87.33%.(4)Effects of hormone concentration on cuttings: The fastest growth rate of plant height is sodium nitrate naphthylacetic acid 250mg/L;The largest number of new leaves on the 15 th and 30 th days was 1200mg/L of ibnaac.On the 15 th day,the largest number of roots was 500 mg/L sodium nitrate naphthylacetic acid;On the 30 th day,the largest number of roots was 1200mg/L indbuconacetic acid.The highest survival rate was 1200mg/L ibnaa,which was 91.33%.(5)Effects of substrate disinfection on cuttings: The fastest growth rate of plant height is 300mg/L methyl sulfur bacteria;On the 15 th day,the largest number of new leaves was 810mg/L carbendazim;On the 30 th day,the largest number of newborn leaves was 300mg/L thiophanate methyl.On the 15 th day,the largest number of roots was 810mg/L carbendazim;On the 30 th day,the largest number of roots was300mg/L thiophanate methyl.The highest survival rate was 300 mg/L thiophanate methyl,which was90.67%.Through the analysis of each processing data,the method to improve the cutting survival of Medicago falcata branches is to select Medicago falcata in budding stage as cutting material,treat the cutting branches with leaves retained with 1200mg/L indanoic acid,use flower soil as cutting matrix and disinfect with 300mg/L thiophanate methyl. |
