| Theileria equi is a parasitic disease that endangers the health of equines.It is caused by the blood protozoiasis Theileria equi and Theileria haneyi.It is spread by ticks in tropical and subtropical regions.Its clinical features are pale mucosa,edema of limbs,jaundice,fever,hepatitis,and splenomegaly.High morbidity and mortality seriously hindered the development of the horse industry in China.In this paper,molecular biology techniques were used to investigate the infection situation of Theileria equi in some areas of Ili,and phylogenetic evolution was used to analyze the genotype of horse Theileria equi in Xinjiang.The differential proteins of equine blood red blood cells before and after infection with horse Theileria equi were screened by proteomic methods,to provide a reference for the epidemic and prevention and control of Theileria equi,to provide a reference for the epidemic prevention and control of Theileria equi.The main experimental contents are as follows:1.In order to know the infection rate of Theileria equi in Ili,this experiment collected A total of302 horse blood samples were collected from 7 experimental sites in 3 counties of Ili,and specific primers were designed based on the 18 S rRNA gene sequence of Theileria equi,and the samples were detected by PCR.The results showed that the infection rate of Theileria equi was 23.8%(72/302).Among the seven experimental sites,the highest infection rate was in the Ka Xia Jiaer area of Zhaosu,with an infection rate of 38%.2.Sequence and identification of the 18 S rRNA gene sequence of Theileria equi were performed.The phylogenetic tree was constructed with Mega software,and the genetic distance was calculated.The results showed that the genotype of Theileria equi in the Ili area of Xinjiang was E,the genetic relationship between genotype B and genotype E was closer.3.The protein expression levels of horse red blood cells before and after infection with Theileria equi were qualitatively and quantitatively analyzed by Label-free proteomics technology.Compared with before infection,a total of 39 differential proteins were identified in horse red blood cells after infection,including 14 up-regulated proteins and 25 down-regulated proteins.The different proteins were all from equine red blood cells,mainly including Annexin,Glutathione synthetase,Eosinophil peroxidase.The GO function analysis and KEGG enrichment analysis of the differential proteins were carried out,and the results showed that the differential proteins were mainly involved in biological processes such as cellular processes,biological regulation,and metabolic processes.In terms of molecular functions,it is mainly manifested as catalytic activity,transport activity,and antioxidant activity. |
