Stripe rust is a serious disease that threatens wheat production.Planting disease-resistant varieties is the most effective measure for preventing and controlling stripe rust.However,the excessive use of disease resistant genes in the cultivation of disease resistant varieties can easily be overcome by new pathogenic bacteria,resulting in the loss of resistance in disease resistant varieties and causing a pandemic.Therefore,diversifying the use of disease resistance genes and screening a series of materials with known disease resistance genes or polygenic aggregates can provide materials for further breeding of durable disease resistant varieties.The adult plant resistance gene Yr48 was introduced into Chuanmai 42(including Yr26)to construct a high-generation selection population of Chuanmai 42/PI 610750,and 10 lines containing Yr48 containing stripe rust resistance and excellent agronomic traits were selected in the F6and F7generations,but no molecular marker detection was performed on Yr26.Based on this,this study conducted molecular marker detection of F9generation population,and selected lines with resistance to stripe rust and excellent agronomic traits containing Yr48 and Yr26+Yr48.At the same time,the monogenic line wheat PI 660057 contains a high-temperature adult plant resistance gene Yr52,the monogenic line wheat PI 660061contains a high-temperature adult plant resistance gene Yr59,and the Zhou 8425B contains an all stage resistance gene Yr ZH84 were hybridized,and then hybridized with the main cultivated varieties Xikemai 18,Chuanmai 802,Mianmai 367,and Mianmai 902 to construct an F2segregating population.Selected a series of known disease resistance genes or polygenic polymeric materials.The main findings are as follows:1.The wheat institute of Southwest University of Science and Technology constructed Chuanmai 42/PI 610750 high-generation selection population(F8,F9)has 62 lines,through the investigation of stripe rust resistance in the planting stage in more than two years from2021-2022,combined with molecular markers,it was detected that 21 lines only contained Yr48(IT=2-6,DS=10%-40%),8 lines only contained Yr26(IT=3,5-7,DS=30%-45%),and15 lines contained Yr26+Yr48(IT=2-4,DS=10%-25%).Combined with the survey of agronomic traits,13 lines with good resistance and excellent agronomic traits were selected,including 6 lines with Yr26+Yr48.2.There are 27 materials in the F2population of Xikemai 18//PI 660061/PI 660057.Through field resistance identification,materials with moderate resistance and above accounted for14.8%.Molecular markers detected 15 materials with Yr52 or Yr59,and one material with high resistance was selected as subsequent breeding material.3.There are 76 materials in the F2population of Chuanmai 802//PI 660061/PI 660057.Through field disease resistance identification,materials with moderate resistance and above accounted for 88.2%.Molecular markers have detected 42 materials containing Yr52 or Yr59,and 11 materials with high resistance was selected as subsequent breeding material.4.There are 17 materials in the F2population of Mianmai 367/PI 660061/PI 660057.Through field resistance identification,materials with moderate resistance and above accounted for 29.4%.Molecular markers have detected 10 materials containing Yr52 or Yr59,and 3 materials with high resistance was selected as subsequent breeding material.5.There are 55 materials in the F2population of Xikemai 18//Zhou 8425B/PI 660061.Through field resistance identification,materials with moderate resistance and above accounted for 50.9%.Molecular markers have detected 6 materials containing Yr59 and 11materials containing Yr ZH84.6.There are 71 materials in the F2population of Chuanmai 802//Zhou 8425B/PI 660057.Through field disease resistance identification,materials with moderate resistance and above accounted for 74.6%.Molecular markers have detected 12 materials containing only Yr52,11 materials containing only Yr ZH84,and 2 materials with Yr52+Yr ZH84.4 materials with high resistance was selected as subsequent breeding material.7.There are 39 materials in the F2population of Mianmai 902/Zhou 8425B/PI 660057.Through field disease resistance identification,materials with moderate resistance and above accounted for 79.4%.Molecular markers have detected 15 materials containing only Yr52,15 materials containing only Yr ZH84,and 6 materials containing Yr52+Yr ZH84.11materials with high resistance was selected as subsequent breeding material.8.Comparing the resistance to stripe rust between the polymerized genes materials and those with single gene,the materials with Yr26+Yr48,Yr52+Yr ZH84 showed stronger resistance than those with single gene. |
