| In this study,GWAS association analysis was used to find the genetic variation loci leading to the difference in salt tolerance of 224 Tartary buckwheat samples.Finally,two genes were screened on chromosome 2 of buckwheat,named FtAUR3 and FtCISIN.FtAUR3 and FtCISIN genes were overexpressed in hairy roots and Arabidopsis thaliana to explore their salt tolerance.The salt tolerance of 224 Tartary buckwheat cultivars was investigated using germination rate and root length as indexes.FtAUR3 and FtCISIN genes were screened out by GWAS correlation analysis and q RT-PCR analysis.In this study,FtAUR3 and FtCISIN genes were cloned from Chuanqiao No.1.Bioinformatics analysis of FtAUR3 and FtCISIN genes showed that the secondary structure of FtAUR3 and FtCISIN genes were random curly,and the protein types were hydrophobic proteins.In order to study the salt tolerance of FtAUR3 and FtCISIN genes,first of all,FtAUR3-1302 and FtCISIN-1302 hairy roots were overexpressed with Methyl Jasmonate(Me JA)and Na Cl treatment.q RT-PCR showed that the expression levels of FtAUR3 and FtCISIN genes were significantly induced by Me JA and Na Cl.Tissue specific expression analysis:Both FtAUR3 and FtCISIN genes were luxury genes with the highest expression levels in stems.Secondly,in arabidopsis thaliana overexpressed FtAUR3-1302 and FtCISIN-1302,the overexpression of FtAUR3 and FtCISIN genes could improve the tolerance of arabidopsis thaliana to salt treatment during germination.q RT-PCR analysis showed that the overexpression of FtAUR3 and FtCISIN genes not only regulated the expression levels of salt stress related genes(SOS3,AVP1,RCD1,CBIL),but also regulated cinnamate-4 in flavonoid synthase related pathways.The expression levels of hydroxylase(C4H),Chalcone isomerase(CHI),flavonoid 3 â-hydroxylase(Fâ3H);Physiological and biochemical analysis showed that the overexpression of FtAUR3-1302 and FtCISIN-1302 decreased the content of malondialdehyde(MDA)and increased the activities of catalase(CAT)and peroxidase(POD).It is suggested that FtAUR3 and FtCISIN may enhance salt tolerance of Tartary buckwheat through jasmonic acid pathway to promote flavonoid and salt stress response related pathways. |
