Wheat stripe rust,caused by Puccinia striiformis f.sp.tritici(Pst),is a main disease seriously threatening the safety of wheat production.Xiaoyan 6(XY6)is a representative cultivar with high-temperature all-stage resistance to Pst,and has the characteristics of non-race-specific and durable resistance.Our research group has sequenced and analyzed the transcriptomes of wheat inoculated with Pst treated under normal temperature(N)and normal temperature transferred to high temperature(NHN),it was found that the expression of small G protein gene Ta RABH1b L was up-regulated by 6.34 times after dual induction by high-temperature and Pst,suggesting that Ta RABH1b L might be involved in the high-temperature resistance of XY6 to Pst.The mainly results of this study are as following:1.Ta RABH1b L functions as a positive regulatory factor in the high-temperature resistance of XY6 to Pst.It was found that the number of pustules per unit area in Ta RABH1b L-scienced wheat leaves under NHN treatment was about 2.61 times higher than that in non-silenced wheat leaves,and silencing Ta RABH1b L significantly increased the length of uredinia and the hyphal density.2.Yeast-two-hybrid(Y2H)and pull-down assays comfirmed the interaction between Ta RABH1b L and transcription factor Ta ERF1L,and bimolecular fluorescent complementation(Bi FC)assay further proved that the two proteins interacted in the nucleus.Two site-specific mutations of Ta RABH1b L,GTP-state Ta RABH1b LQ69L and GDP-state Ta RABH1b LN122I,were obtained.Y2H,Bi FC and pull-down assays showed that Ta ERF1L only interacted with the GTP-state Ta RABH1b LQ69L.3.Silencing Ta ERF1L significantly increased the length of hyphae and the number of haustorial mother cells,and the number of pustules per unit area in Ta ERF1L-silenced wheat leaves under NHN treatment was about 2.10 times higher than that in non-silenced wheat leaves,which decreased the high-temperature resistance of wheat.
