Sheep is one of the earliest domesticated animals,and China is a big country of sheep,sheep and goats are extremely rich in genetic resources.With the development of social economy,the breeding industry of sheep and goats in China has been developing rapidly,and various breeding technology levels and breeding capabilities have been continuously enhanced.The development of animal molecular breeding has made molecular marker technology widely used in sheep and goat breeding.InDel markers have a large number and simple typing,and have been widely studied in animal breeding work,providing a scientific basis for improving animal economic traits.The Hippo signaling pathway is a highly conserved pathway in mammals.It regulates a variety of biological processes,participates in tissue growth and regeneration,regulates cell proliferation and apoptosis,and controls organ size.It plays a very important role in animal growth and development.MST1/2,LATS1/2,SAV1,MOB1A/B and YAP/TAZ genes are important components of the Hippo signaling pathway.MST1/2,LATS1/2,SAV1,MOB1A/B are the core kinases in the pathway,and YAP/TAZ is a key effector in the pathway.In this study,sheep(Tong Sheep,Hu Sheep,Small Tail Han Sheep,Lanzhou Dawei Sheep)and goats(Shanbei White Cashmere Goat,Guanzhong Dairy Goat)were used as research objects.The public database was used to screen the potential InDel in MST1/2,LATS1/2,SAV1,MOB1A/B and YAP/TAZ genes in the Hippo signaling pathway of sheep and goats.The InDel in sheep and goats was identified by designing specific primers,PCR amplification,agarose gel electrophoresis typing,gene sequencing and other techniques,and the key gene InDel in the Hippo signaling pathway of sheep and goats was compared and analyzed.The association analysis of InDel of these genes with growth traits of sheep and goat was carried out,and the key In affecting growth and development was excavated.The results show that :1.There are 154 potential InDel sites in the key genes of sheep Hippo signaling pathway,and 25 InDel sites can be identified in four sheep breeds,including four in MST1 gene,eight in MST2 gene,eight in YAP gene,two in TAZ gene,three in MOB1 A gene,and four in MOB1 B,SAV1,LATS1 and LATS2 genes.Three loci in the MST1 gene were related to the growth traits of Small Tail Han sheep,Hu sheep and Tong sheep,which significantly affected chest circumference,body height,cannon circumference,body weight and other traits.Five loci in the MST2 gene significantly affected the growth traits of cross height,body weight and chest circumference of four sheep breeds.Two loci in the YAP gene significantly affected the body height,chest circumference and body length of Lanzhou Large Tail sheep,Small Tail Han sheep and Tong sheep.Three loci in MOB1 A had the most significant effect on the cannon circumference of four sheep breeds.These 13 InDel loci were closely related to the growth traits of Lanzhou Large Tail sheep,Small Tail Han sheep,Hu sheep and Tong sheep.It can be used as an effective molecular marker for sheep molecular breeding.2.A total of 152 potential InDel loci were screened out in the key genes of Hippo signaling pathway in goats.A total of 25 InDel loci were identified in Shanbei white cashmere goats and Guanzhong dairy goats,including eight in YAP gene,nine in TAZ gene,five in MOB1 B gene,one in SAV1 gene and two in LATS2 gene,while no InDel loci were identified in MST1,MST2,MOB1 A and LATS1 genes.The association analysis between InDel loci and growth traits identified in Shanbei white cashmere goats and Guanzhong dairy goats showed that five loci in YAP gene significantly affected chest depth,chest circumference and cannon circumference.Three loci in TAZ gene were significantly correlated with body height,body length and body weight.The SAV1-G5 loci of SAV1 gene were significantly correlated with chest circumference and cannon circumference.A total of nine InDel loci were closely related to the growth traits of Shanbei white cashmere goats and Guanzhong dairy goats,which could be used as effective molecular markers for molecular breeding of goats.3.The length of InDel identified in sheep and goat was mainly between 6-10 bp,and the longest was 27 bp;in addition,the number of deletion mutations in the identified InDel locus was more than that of insertion mutations.MST1,MST2 and MOB1 A genes identified InDel only in sheep,MOB1 B,SAV1 and LATS2 genes identified InDel only in goats,YAP gene identified more InDel in sheep and goats,and the number of InDel loci identified in TAZ gene of sheep was much less than that identified in goats.In summary,the InDel of MST1/2,LATS1/2,SAV1,MOB1A/B and YAP/TAZ genes in the Hippo signaling pathway have similarities and differences between sheep and goats,and are closely related to the growth traits of sheep and goats.They can be used as candidate genes for molecular breeding of sheep and goats.Thirteen InDel loci in sheep and nine InDel loci in goats can be used as InDel molecular markers for the breeding of excellent breeds.
