| Vitamin D receptor is essentially a ligand-dependent nuclear transcription factor,which plays an important role in maintaining calcium-phosphorus metabolism and regulating cell proliferation and differentiation.Hair follicle is a special appendage of cashmere goat skin,which is located in the dermis of the skin and occurs in the embryonic stage of cashmere goat.Due to the signal molecular transmission between dermal cells and epithelial cells,its development changes periodically,which goes through anagen,catagen and telogen.In this study,on the basis of high-throughput transcriptome sequencing data in the laboratory,VDR gene expressed differentially in the growth cycles of Albas white cashmere goat hair follicles were obtained,and its expression is detected at the gene level,protein level,and tissue site by q RT-PCR,Western blot,and immunohistochemistry techniques.The miRNA and circ RNA regulating VDR gene were predicted by bioinformatics method and identified by double Luciferase reporting experiment.The results are as follows:1.There are significant differences in the expression of VDR and its genes i n the growth cycles of cashmere goat hair follicles.The expression level in anage n is significantly higher than that in telogen,followed by that in catagen.There a re significant differences in the expression of VDR and its genes in the hair follic le growth cycle of cashmere goats.The expression in the anagen was significantly higher than that in the telogen,followed by that in the catagen.2.VDR is expressed in outer root sheath,inner root sheath and hair matrix of primary and secondary hair follicles of Albas white cashmere goats,and the ex pression level of hair matrix is the most.3.MiR-124 b is a potential miRNA regulating VDR gene.The expression trend of miR-124 b and VDR gene in the hair follicle growth cycles is opposite.The dual Luciferase reporting system verified that miR-124 b has inhibitory effect on target VDR gene.4.Dual luciferase reporter system showed that circ FLVCR1-005 competitively binds miR-124 b to promote VDR gene expression.The purpose of this study is to lay a theoretical foundation for the follow-up study on the periodic growth and development mechanism of secondary hair follicles and the screening of molecular targets. |
