Study On The Effect Of Dietary Suplementation Of Lasia Spinosa Powder On The Spermatogenesis Of Mice And Landrace Boars

Abstract/Summary:
The spermatogenic function of male breeding animals often declines because of heavy breeding tasks,poor breeding management,environmental mutation and other factors.Lasia spinosa is a kind of herb with antioxidant,antibacterial,anti-inflammatory and hormone-like activities,and it has been shown that Lasia spinosa can promote spermatogenesis in old roosters.However,the effect of Lasia spinosa on spermatogenesis in male mammals remains unclear.Therefore,the following two experiments were conducted in this study to explore the effects of Lasia spinosa on spermatogenesis in male mammals.Experiment 1: The diets of 4-week-old male mice were supplemented with Lasia spinosa powder,and the effects of Lasia spinosa powder on body weight,testis coefficient,lumen diameter of testis flexion fine duct,testis antioxidant,testis gene expression level,semen quality,serum reproductive hormone and biochemical index of the mice were investigated after 60 days of feeding.The results showed that there was no significant difference in body weight between the experimental group and the control group(P>0.05),but the testicular coefficient and the lumen diameter of the testicular curvature were significantly increased(P<0.05).Testis antioxidant activity: Lasia spinosa powder could decreased the level of reactive oxygen species(ROS),increased the concentration of glutathione(GSH)and the activity of total superoxide dismutase(T-SOD)in testis tissue,all of which were significant(P<0.05).Semen quality: Semen count,sperm motility,sperm motility and sperm motility of the epididymis were significantly increased(P<0.05),and sperm malformation rate was significantly decreased(P<0.05).Semen quality of 2%test group was the best.Reproductive hormone levels: serum concentrations of testosterone(T),luteinizing hormone(LH)and follicle-stimulating hormone(FSH)in experimental group were not significantly different from those in control group(P>0.05).Serum biochemical level: Lasia spinosa powder could significantly reduce serum triglyceride concentration and increase serum calcium and phosphorus ion concentration(P<0.05),but there were no significant differences in other biochemical indexes between experimental group and control group(P>0.05).A total of 531 differentially expressed genes were identified in the test group and the control group,of which 311 were down-regulated and 220 were up-regulated.Experiment 2: The diet of 36-month-old Landrace Boars was supplemented with Lasia spinosa powder to investigate the effects of Lasia spinosa powder on semen intake,semen quality,seminal fructose concentration,serum biochemical index and mating potency of boar were investigated after 90 days of feeding.The results showed that there was no significant difference in sperm collection amount between the experimental group and the control group(P>0.05),but the sperm density,sperm motility and sperm motility of boar were significantly increased,and the sperm malformation rate was decreased(P< 0.05).Lasia spinosa powder could improve the mating quantity of boar semen,but there was no significant difference in mating results between experimental group and control group(P>0.05).There was no significant difference in semen fructose concentration between experimental group and control group(P>0.05).Lasia spinosa powder significantly decreased the activity of alanine aminotransferase(ALT)and increased the concentration of calcium ions in serum of boars(P<0.05),but there were no significant differences in other biochemical indexes between experimental group and control group(P> 0.05).In summary:(1)The spermatogenic function,testis coefficient and antioxidant capacity of mice and semen quality of mice were enhanced by adding Lasia spinosa powder to the diet,but the body weight and body health of mice were not significantly affected,and the proportion of adding 2% was the best.(2)Adding Lasia spinosa powder to the diet can improve semen quality and mating efficiency of boars,but has no significant effect on the health status of boars.The results of this study provide theoretical and practical reference for the application of Lasia spinosa powder as plant feed additive in the production of animal feed.
