Hu sheep is an excellent local sheep breed in China and Hu sheep meat have tender and fresh and rich nutrition characteristic.With the improvement of people’s living standards,people pay more and more attention to food safety.Mogrosides is an extract of Grosvenoside,which has the effects of treating diabetes,lowering blood lipid,anti-inflammation,liver protection and anti-fibrosis.However,it has been less studied in ruminant production applications.Therefore,this study aims to explore the effect and mechanism of adding of Mogrosides to the diet on the production performance and mechanism of Hu sheep.This will provide experimental basis for the development of Mogrosides as feed additive to improve the production efficiency of ruminants.1.Forty-five 7-month-old Hu sheeps were randomly divided into three groups(control group,900 mg/kg Mogrosides group and 1200 mg/kg Mogrosides group).And,the slaughter performance and meat quality were determined during the formal feeding period of 56 days.The 900 mg/kg Mogrosides and 1200 mg/kg Mogrosides can significantly increase the feed intake(P<0.05),accelerated the growth rate(P<0.05),decreased the feed-to-weight ratio(P<0.05),and increased carcass weight and net meat weight(P<0.05)compared with the control group.And,the 900 mg/kg Mogrosides and 1200 mg/kg Mogrosides group significantly increased the redness of meat color(a*)and intramuscular fat content(P<0.05),reduced the shear force(P<0.05).In summary,Mogrosides can improve the growth performance and meat quality of Hu sheep.2.The gene expression of the longest dorsal muscle of 1200 mg/kg Mogrosides group and the control group was analyze by RNA-seq.It was found that a total of 267 differentially expressed genes were screened for the dorsal muscle on 1200 mg/kg Mogrosides group(log2| Fold Change|>1,P<0.05)(including 135 up-regulated differential genes(SCL16A12,WNT4,DGAT2,etc.)and 132 down-regulated differential genes(FOS,THRSP,PCK2,etc.)compared with the control group.The differential expressed gene mainly annotated the MAPK and Wnt related to muscle development signaling pathway and PPAR signaling pathway,cytochrome P450,fat digestion and absorption related to fat metabolism pathways.3.The rumen microorganism structure of the 1200 mg/kg Mogrosides group and the control group were analysis by 16 S r DNA sequencing.It showed that the dominant phylums of 1200 mg/kg Mogrosides group are Bacteroides,Firmicutes and Proteobacteria and the dominant genus were Peurella,Phyceticaceae_RC9_gut_group,and Succinic acidobacteria.And,the relative abundance of Bacteroides phylum increased significantly(P<0.05)and the relative abundance of Firmicutes phylum decreased significantly(P<0.05)in 1200 mg/kg Mogrosides group compare with control group.Functional predictive analysis found that the differential microorganisms were significantly enriched pathways of lactam biosynthesis,glycolysis/gluconeogenesis,glycosaminoglycan degradation,fat digestion and absorption,and apoptosis in 1200 mg/kg Mogrosides group.The control group was mainly enriched in ABC transporter,cytochrome P450 metabolism of exogenous substances and endoplasmic reticulum protein processing.In summary,Mogrosides-supplemented diet can enhance the absorpti on and metabolism of nutrients,improve the diversity of rumen flora,in crease the expression of genes related to muscle development to improve the growth performance and meat quality of Hu sheep.
