| Platanus L.belongs to the Platanaceae family and has high garden application value.Many researchers are committed to germplasm innovation and excellent variety selection.At present,several new varieties of sycamore have been approved and authorized one after another,but there are no basic documents for testing new varieties of sycamore.Therefore,it is crucial to develop DUS testing guidelines for Platanus L.,which can provide technical support and standardized specifications for the testing and judgment of new varieties of sycamore.In addition,with the promotion and application of new varieties of sycamore,the protection of their related rights and interests has also emerged.Based on previous research,this study further conducted on-site investigations on the germplasm resources of Platanus L.conducted specific analysis and grading of test traits,constructed a database of known Platanus L.varieties,screened and optimized the test traits and standard varieties of Platanus L.DUS testing guidelines.By consulting information and combining relevant documents,a reliable and effective DUS testing guide for new Platanus L.varieties was further developed;In addition,based on SSR and ISSR molecular marker technologies,the construction of molecular fingerprints and the development of new molecular markers were carried out.The main research results are as follows:(1)A field investigation was conducted on the included varieties of sycamore,and there were more varieties surveyed than included.However,due to the lack of approval and authorization for new varieties and the difficulty in accessing textual information on other reported varieties,a total of 13 varieties of sycamore were included.Through on-site investigation and analysis of the phenotypic traits of Platanus L.,adjustments and additions were made to the 19 mandatory traits identified by previous studies,and 26 DUS test traits were identified,including 6 quality traits,accounting for 23.07%;11 quantitative traits,accounting for 42.31%,and 9 pseudo quality traits,accounting for 34.61%;23 mandatory traits,3 auxiliary test traits,and 15 graphical traits.(2)Using SSR molecular markers to amplify 18 samples of Platanus L.,a total of 13 pairs of primers were screened out,and 146 bands were amplified,with 141 polymorphic bands.The polymorphism ratio of each primer pair was between 90.91% and 100.00%;Translate the variety information and fingerprint maps of 18 varieties of sycamore trees into fingerprint QR codes for their promotion and application.(3)Based on ISSR molecular marker technology,specific bands were transformed using SCAR markers.A total of 98 bands were amplified from 12 ISSR markers in 18 new varieties of Platanus L.,with 85 polymorphic bands.The polymorphism ratio of each primer pair ranged from 75.00% to 100.00%.There are 2 pairs of primers that can obtain specific bands,which can temporarily distinguish the varieties of ‘P.acerifolia Susheng’,‘P.acerifolia Xiongfeng’,and ‘F27’ from the 18 varieties of sycamore. |
