Objective:We can study the Chinese medicine prescription of Tonifying kidney and excreting dampness method to prevent the mechanism of Gouty Arthitis(GA)by the expression level of IL-17、substance P and COL-Ⅰ.It is that the research on the regulation of GA inflammatory response and bone destruction in rats by Tonifying kidney and excreting dampness method,which used to provide new ideas and methods of the prevention of gouty arthitis.Method:84 SD rats were randomly divided into seven groups,respectively they were blank group(BG),model group(MG),the control group of gout relief(GRG),colchicine group(CG),low-dose、middle-dose and high-dose group of Chinese medicine prescription of Tonifying kidney and excreting dampness method(CMLG、CMMG、CMHG)(N=12).Complexed model using oxygen oxazine acid potassium and sodium urate forming new method induced experimental model,namely a rat model of gouty arthritis.We used physiological saline to gavage normal control group and model group,while used Tongfeng Shu solution to gavage the GRG and used colchicine solution to gavage the CG.Then we used low-dose、middle-dose and high-dose of Chinese medicine prescription of Tonifying kidney and excreting dampness method to gavage the corresponding group with the administration in terms of the proportion of measurement followed by 5ml·kg-1、10ml·kg-1、20ml·kg-1.Various groups were gavaged by 1 mL with 100g body weight of rats as a standard,once daily.After 7 days later,we observed the level of IL-17 and SP of the serum of every groups,and the level of COL-Ⅰ in the synovial tissue.Result:The impaction of the serum level of IL-17 and substance P and the expression of COL-Ⅰ in the synovial tissue from acute gouty arthritis model rats of all groups was conclued to parts.Firstly,compared with the BG group,the serum level of IL-17 and substance P of MG increased obviously(P<0.05),and the expression of COL-Ⅰ in the synovial tissue of MG increased,too(P<0.05).Secondly,except the BG,compared with the MG,the serum level of IL-17、substance P and the expression of COL-Ⅰ in the synovial tissue of the other groups all decreased(P<0.05).Thirdly,compared with GRG,the serum level of IL-17、substance P of CG and CMMG decreased obviously(P<0.05),and the expression of COL-Ⅰ in the synovial tissue has a little bit lower.Besides,the expression of COL-Ⅰ in the synovial tissue of CMHG decreased obviously.In addition,the expression of COL-Ⅰ in the synovial tissue of GA rats reduced obviously among the CMLG、CMMG and CMHG,and the most lowest group is CMHG.Conclution:The experiment proved that IL-17、Substance P and COL-Ⅰ participate in the procession of the inflammatory response of GA rat for the first time and the prescription of Tonifying kidney and excreting dampness method can reduce the content of serum IL-17,substance P and COL-Ⅰ in synovial tissue of GA rat at the same time.The prescription of Tonifying kidney and excreting dampness method in middle-dose can reduce the serum IL-17,substance P of GA rat obviously,while the prescription of Tonifying kidney and excreting dampness method in high-dose is sensitive to the expression of COL-Ⅰ.
