| Objective In this research,we,based on the theory of traditional Chinese medicine and modern medicine,use different doses of Erguotou to intervene in the model rats of nonalcoholic fatty liver disease(NAFLD)induced by high fat diet.Then,We observe the general condition and pathological changes in liver tissue of each group of rats and detect the liver function,serum lipid and TGF-β1,CollagenI,α-SMA mRNA in liver tissue.We observe and analyze the changes of them.Especially,the effects of different alcohol consumption to the expression of TGF-β1,CollagenI and α-SMA mRNA in liver tissue and the relationship with the occurrence and development of liver fibrosis to further explore the pathogenesis of NAFLD,Looking forward to provide guidance for safe drinking in patients with clinically nonalcoholic fatty liver disease.Methods After adaptive feeding for a week,forty SD male rats were randomly divided into four groups:normal control group,NAFLD model group,moderate drinking group,excessive drinking group.The rats in the normal control group were fed with standard diet and the other groups were fed with high fat diet.After 8 weeks.Rats of moderate drinking group and excessive drinking group begin to fed with Erguotou by gavage,the alcohol dose was 2.0g/kg· d,and 4.0/Kg·d respectively.At the same time,normal control group and NAFLD model group were fed with equal volume of saline by gavage.After 17 weeks,when all rats were anesthetized with chloral hydrate,we take the blood and liver tissue of rat,detect the level of serumTG,TC,HDL,LDL,ALT,AST,by automatic biochemical analyzer and the expression of TGF-β1,CollagenI and α-SMA genes in liver tissue by PCR,observe the histological changes of fatty steatosis,inflammation and fibrosis in liver tissue of rats in each group through oil red,HE,masson staining method under the microscope.Finally,we anlyze the changes of general situation,serum,histology and gene expression between groups.Results(1)Compared with the normal control group,the level of liver index,serum ALT,AST,ALP,LDL-C,TC and the expression of TGF-β1,α-SMA mRNA liver tissue of NAFLD model group significantly rises(P<0.01).The level of serum TG in liver tissue rise and the difference was not statistically significant(P>0.05).The expression of collagen Ⅰ mRNA in liver tissue rises and the difference was statistically significant(P<0.05).(2)Compared with NAFLD model group,the serum liver index of moderate drinking group and the difference was significant(P<0.01).The levels of serum ALT,AST,ALP,LDL-C and TG drops and the difference was statistically significant(P<0.05).The expression of serum TGF-β1,α-SMA and Collagen Ⅰ mRNA in liver tissue drops,but the difference was not statistically significant(P>0.05).The level of serum HDL-C rises(P>0.05)and the difference was not statistically significant;(3)Compared with NAFLD model group,the level of serum ALP,LDL-C,TC in excessive drinking group rises and the difference was significant(P<0.01).The level of liver index,serum ALT and AST were rises(P<0.05).The expression of TGF-β1,α-SMA and Collagen Ⅰ mRNA in liver tissue and level of serum TG rises,but the difference was not statistically significant(P>0.05).Conclusion The effect of alcohol consumption on the expression of TGF-β1,α-SMA and CollagenⅠ mRNA in liver tissue of NAFLD model is related to the dose.Moderate drinking can inhibit th e expression of TGF-β1,α-SMA and CollagenⅠ mRNA in the liver to prevent the occurrence and development of liver fibrosis in NAFLD.Excessive drinking can increase the expression of TGF-β1,α-SMA and CollagenⅠ mRNA in the liver tissue to promote the ccurrence and development of liver fibrosis in NAFLD. |
