Study On The Effect Of Xiao’er Jianpi Plaster And Its Components On Gastrointestinal Motility

Objective By orthogonal design method,to study the effect of Xiao’er Jianpi Plaster on gastric and ileal smooth muscle of normal rats in vitro,as to compare the contribution of each component to the gastrointestinal motility of the whole recipe,to select the optimum concentration ratio;And to investigate the effect of the whole party and disassembly of the Xiao’er Jianpi Plaster on gastric and ileal smooth muscle of normal rats in vitro,to identify the drug and its compatibility in the prescription of gastrointestinal motility,so as to lay a foundation for the study of its effective substances.Methods The biological signal acquisition system was used to record the changes of the tension of the normal gastric and ileal smooth muscle of the rats,and to calculate the percentage change.Results ①By orthogonal design method,the contribution of each component to the contraction of gastric smooth muscle:A(clove)>C(evodia)>B(hawthorn)>D(cinnamon),E(white pepper)had no significant effect on the contraction of gastric smooth muscle;The contribution of each component to the contraction of ileal smooth muscle:A(clove)>B(hawthorn)>C(evodia),D(cinnamon)and E(white pepper)had no significant effects on the contraction of ileal smooth muscle;The optimal proportion of the Xiao’er Jianpi Plaster was selected:high dose level of clove,high dose of hawthorn,small dose of evodia,high dose of cinnamon,low dose of white pepper.②The effect of the whole recipe and its components on the contraction of gastric smooth muscle in rats:the whole recipe>clove>hawthorn>cinnamon,the effect of white pepper was not significant;The effect on the contraction of ileal smooth muscle in rats:clove>the whole recipe>hawthorn,the effects of cinnamon and white pepper were not significant,evodia had a bidirectional regulation of the gastrointestinal contraction,it could promote in the small dose,while the dose increased,it had an inhibitory effect.③The effect of the whole recipe and its components on the contraction of gastric smooth muscle in guinea pigs:clove>the whole recipe>evodia>hawthorn,the effects of cinnamon and white pepper were not significant;The effect on the contraction of ileal smooth muscle in guinea pigs:clove>the whole recipe>hawthorn,evodia had an inhibitory effect,and the effects of cinnamon and white pepper were not significant.The synergistic effect of the two compatibility on the contraction of gastric smooth muscle in guinea pigs:clove+cinnamon,hawthorn+cinnamon,evodia+cinnamon and clove+white pepper;The synergistic effect of the contraction of ileal smooth muscle:clove+cinnamon,clove+ white pepper,hawthorn+cinnamon;however,evodia+cinnamon and evodia+white pepper showed inhibitory effects.Conclusion ①The Xiao’er Jianpi Plaster can promote the gastrointestinal motility.②In Xiao’er Jianpi Plaster recipe,clove and hawthorn have significant promoting effects on the gastric and ileal smooth muscle in vitro,while evodia has a bidirectional regulation;clove and hawthorn are the main ingredients that promote the gastrointestinal motility in this prescription.③The effect of gastrointestinal motility in the Xiao’er Jianpi Plaster is not a simple addition and subtraction of each single drug or its disassembling effect,but it’s an organic integration,which reflects the complexity and applicability of the combination of TCM compatibility.
