As a protective shield for the body from the external environment,the skin is constantly exposed to potential injury.After injury,the skin or other organs has self-repair ability.Wound healing is a complex and highly coordinated biological process.This process involves a variety of cytokines,reparative cells and extracellular matrix,which interact with each other.Wound healing encompasses spatially and temporally overlapping processes including blood clotting,inflammation and cellular proliferation and extracellular matrix(ECM)remodeling.Angiogenesis makes possible the re-supply of oxygen and other nutrients.The key to promote wound healing is the regulator of angiogenesis.It has been confirmed that some growth factors,proteins,traditional Chinese medicine can regulate angiogenesis.In this paper,the role of vascular regeneration in wound healing is discussed from two aspects:growth factor and traditional Chinese medicine.1.Activin B promotes the regeneration of blood vessels during the skin wound healing(1)Background and ObjectiveThe key to promote wound healing is the regulator of angiogenesis.Currently,studies have shown that TGF-β plays an important role in angiogenesis.Our group early findings that activin B promotes the proliferation and migration of keratinocytes around wound,which can accelerate wound healing and promote re-epithelization.Activin B is the one of transforming growth factor-β superfamily members.The effect of activin B on the blood vessels regeneration is unclear.Our objective is to elucidate the role of activin B in blood vessels regeneration during skin wound healing.In vitro,Our objective is to explore activin B participates in angiogenesis by the regulator of HDMECs.Thus,it provides a possible research basis for the application of Activin B in skin wound healing.(2)MethodsThe full-thickness skin excision model of mice was used.The wounds in treatment group were locally treated with activin B,the wounds in the control group with PBS respectively.Wound closure rate was measured.The formation of granulation tissue was observed.The blood vessels regeneration was analyzed by immunohistochemistry.The blood flow in wound granulation tissue was detected by skin micro-circulation testing equipment.In vitro,the cell proliferation activity was determined using cell counting Kit-8(CCK-8)assay and EdU assay.Wound healing and trasnswell assay were used to assess the migration and chemotaxis of HDMECs,respectively.The level of TGF-β1 and VEGF in supernatant fluid of fibroblasts treated with activin B was detected by double antibody sandwich enzyme-linked immunosorbent assay(ELISA).(3)ResultsAfter the application of activin B in skin wound,the rate of wound healing was accelerated.The number of revascularization in wound granulation tissue increased,and the average blood flow of granulation tissue was higher.Activin B was unable to regulate the cell proliferation activity,the migration and chemotaxis of HDMECs.The level of VEGF in supernatant fluid of fibroblasts treated with activin B was high.(4)ConclusionActivin B can promote the regeneration of blood vessels in wound granulation tissue and accelerate wound healing.In vitro,the high expression of VEGF in supernatant fluid of fibroblasts treated with activin B can promote the proliferation of HDMECs.2.Glycyrrhiza promotes the regeneration of blood vessels during the skin wound healing(1)Background and ObjectiveScald is a common and extremely complex trauma in daily life.In modern medicine,all kinds of traditional Chinese medicine(TCM)play a very important role in the treatment of mild or severe scald.Glycyrrhizae,as TCM,has antibacterial,Anti-inflammatory,analgesic effects.Our objective is to elucidate the role of glycyrrhizae in wound healing and in blood vessels regeneration after deep Ⅱ degree scald injury in mice.(2)MethodsThe mice were randomly divided into four groups,namely,control group(normal healing),negative control(sesame oil),positive control(MEBO)and experimental group(Glycyrrhizae)after deep Ⅱ degree scald injury in mice.Wound closure rate was measured.The formation of granulation tissue was observed.The blood vessels regeneration was analyzed by immunohistochemistry.The blood flow in wound granulation tissue was detected by skin micro-circulation testing equipment.Collagen synthesis was observed using Masson trichrome staining.(3)ResultsAfter the application of Glycyrrhizae in skin wound,compared to the other three groups,the rate of wound healing was accelerated.The number of revascularization in wound granulation tissue was increased,and the average blood flow of granulation tissue was higher.(4)ConclusionGlycyrrhizae can promote the regeneration of blood vessels in wound granulation tissue and accelerate wound healing.
