| Background:Ulcerative colitis(UC)belongs to the category of Inflammatory bowel disease(IBD).Clinically,it was found that the proportion of patients with UC accompanied by depressive state is relatively high,especially in the active stage of UC disease.UC is often accompanied by gut microbiota disorder.Microbiota-gut-brain axisdysfunction has become an important pathogenesis of depression.It is necessary to further explore the correlation between UC-microbiota-depression in active stage.Methods:This study included 129 active UC patients(75 with depression and 54 without depression),49 non-UC depressive patients and 62 healthy persons.16srRNA method was used to detect gut microbiota of all subjects,and non-targeted serum metabolomics was used for multi-group association analysis to identify gut microbiota and serum metabolomics characteristics related to UC depression.Results:Active UC patients with depression have unique intestinal microecology andserum metabolomics.Compared with UC without depression,the gut microbiota of UC patients with depression such as Sellimonas and Anaerofustis is increased.Prevotella9、Eubacteriumrectalegroup、Paracoccus、Petrimonas、Ruminococcusgauvreauiigroup、Cellvibrio、Pseudobutyrivibrio、Thermobifida and other bacteria decreased.Sellimonas、Anaerofustis and Prevotella9 three UC-depressive comorbidities were found.The content of me-tabolites such as 1-Stearoyl-sn-glycerol、1-Stearoyl-rac-glycerol、1-O-(cis-9-Octadecenyl)-2-O-acetyl-sn-glycero-3-phosphocholine and Glycochenodeoxycholate increased,and the content of metabolites such as 4-hydroxide benzoate,gamma-l-glutamyl-l-glutamic acid,2’deoxyy-d-ribose decreased in serum of UC patients with depression.T-here is a strong correlation between UC depression core metabolites such as Prevotella9、Eubacteriumrectalegroup、Ruminococcusgauvreauiigroup、Cellvibrio、Pseudobutyrivibrio、Thermobifida and UC depression core metabolite such as 1-Stearoyl-sn-glycerol、1-Stearoyl-rac-glyce rol、1-O-(cis-9-Octadecenyl)-2-O-acetyl-sn-glycero-3-phosphocholine.In addition,Mediation analysis found a combination of e.g.Eubacterium ruminantiumgroup,1-Stearoyl-rac-glyce rol metabolite and UC depression,representing an intermediary effect mode in which microbiota indirectly regulates UC depression phenotype through metabolites.Conclusion:UC patients with depression have unique gut microbiota and serum metabolic characteristics.Sellimonas、Anaerofustis and Prevotella9 may play an important role in UCdepression comorbidity.Bacteria-metabolite combinations such as Prevotella9、Eubacteriumrectalegroup、1-Stearoyl-sn-glycerol、1-Stearoyl-rac-glycerol are strongly correlated in UC depression.Eubacteriumruminantiumgroup and other bacteria may indirectly regulate UC depression phenotype through metabolites such as 1-Stearoyl-rac-glycerol.This study believes that dysfunction of gut microbiota-intestine-brain axis may be the cause of UC depression.Further research is needed to confirm the causal relationship between depression and microbiota changes and the mechanism of related bacteria and metabolites in UC depression. |
