| ObjectiveThis study was aimed to explore the effects of evodiamine on the proliferation,cycle distribution and apoptosis of human nasopharyngeal carcinoma CNE-2 cells and the proble mechanism.Methods1.Human nasopharyngeal carcinoma CNE-2 cells were cultured in vitro.2.The inhibitory effect of evodiamine on the proliferation of CNE-2 cells was analyzed by CCK-8 method.3.The morphological changes of CNE-2 cells treated with evodiamine were observed by optical microscope.4.The effect of evodiamine on cell cycle and apoptosis of CNE-2 cells was detected by flow cytometry.5.The effect of evodiamine on MAPK and STAT3 signal pathways in CNE-2 cells was detected by Western Blot.Results1.Evodiamine inhibited the proliferation and decreased the survival rate of CNE-2 cells in a concentration-and time-dependent manner.And the IC50 values of 24 h and 48 h were 3.157 μM and 0.512 μM,respectively.2.With the treatment of evodiamine,the morphology of the CNE-2cells changed.These cells became round,shrunk,smaller and fragmented with the increase of drug concentration,and there were abnormal cells appeared.The number of cells decreased,while the number of floating cells increased.3.Evodiamine induced more CNE-2 cells in G2/M phase in a dose-dependent manner.4.Evodiamine increased the apoptosis rate of CNE-2 cells in dose-and time-dependent manner.5.Evodiamine reduced the expression of p-p38,p-ERK,p-JNK and p-STAT3 in CNE-2 cells.Conclusion1.Evodiamine inhibited the proliferation of CNE-2 cells,induced apoptosis and promoted cell cycle arrest in G2/M phase.2.MAPK and STAT3 signaling pathways mediated by evodiamine might be involving in this produce. |
