Objective:1.To investigate the association of hepcidin with CHD and coronary atherosclerotic severity.2.To explore the correlation between hepcidin and Ox-LDL and whether hepcidin causes atherosclerosis by catalyzing the information of Ox-LDL.3.to analyse whether the levels of hepcidin affects the incidence of CHD in premenopausal and postmenopausal women.Methods:1.In this study,166 patients with coronary heart disease were diagnosed by coronary angiography.The patients were divided into male CHD group of 83 cases and female CHD group of 83 cases,then the female CHD group was divided into premenopausal group of 24 cases and postmenopausal group of 59 cases.At the same time,the control group,with negative coronary angiography,consisted of 36 cases of men and 40 cases of women,then the female control group was also divided into premenopausal group of 18 cases and postmenopausal group of 22 cases.2.The study collected the fasting venous blood samples of subjects.And then they were sent to the laboratory of our hospital to test low-density lipoprotein(LDL),triglyceride(TG),apolipoproteinA1(apoA 1),high-density lipoprotein(HDL),apolipoproteinB(apoB),total cholesterol(TC).The plasma levels of Hep and Ox-LDL were determined by enzyme-linked immunosorbent assay(ELISA)method.At the same time,the general data which included gender,age,blood pressure,historyof smoking and drinking,primary disease,height and weight of the subjects were collected.All cases was performed with coronary angiography and Gensini score by professional physician in our hospital.3.Firstly,the normality of the data will be tested.If the data is distributed in a normal fashion,the independent sample t-test was used for the comparison of the measurement data between the two groups,and the single-factor completely random variance analysis was used for the comparison between the three groups.Mann-whitney U test will be used when the data is not normal.The spearson correlation analysis was used to analyze the correlation of hepcidin with TG,HDL,LDL,TC,apoA1 and apoB.The binary logistic regression analysis was used toanalyze the related influencing factors of CHD.Result:1.The median levels of hepcidin in CHD group and control group were 20.470μg/L,17.599μg/L,respectively,the median levels of Ox-LDL in CHD group and control group were 86.334μg/L,69.879μg/L,respectively,the different levels of hepcidin and Ox-LDL showed a statistical significance between the groups(P<0.05).Logistic multivariate analysis showed that LDL,Ox-LDL and hepcidin were risk factors for CHD.2.The levels of hepcidin of women with CHD was 16.829±4.459μg/L in the premenopausal group and 21.208±6.286μg/L in the postmenopausal group,the different levels of hepcidin showed a statistical significance between the groups(P<0.05).The levels of hepcidin of women with control group in the premenopausal group and the postmenopausal group were 15.603±3.986μg/L,9.203±4.903μg/L,respectively,the different levels of hepcidin showed a statistical significance between the groups(P<0.05).The levels of hepcidin of men with CHD in the group with age greater 50 and group with age not greater 50 were 22.554±6.345μg/L,24.842±8.235μg/L,respectively,the different levels of hepcidin showed no statistical significance betweenthe groups(P>0.05).The levels of hepcidin of men with control group in the group with age greater 50 and the group with age not greater 50 were 18.279±4.592μg/L,18.309±5.725μg/L,respectively,the different levels of hepcidin showed no statistical significance between the groups(P>0.05).3.The median levels of Ox-LDL of women with CHD was 82.180μg/L in the premenopausal group and 98.455μg/L in the postmenopausal group,the different levels of Ox-LDL showed no statistical significance between the groups(P>0.05).The levels of Ox-LDL of women with control group was 81.482±19.444μg/L in premenopausal group and 73.217±20.184μg/L in postmenopausal group,the different levels of Ox-LDL showed no statistical significance between the groups(P>0.05).4.The median levels of hepcidin and Ox-LDL in subjects with normal and low-risk Gensini scores were significantly lower than those with medium-risk and high-risk scores(P<0.05).In the correlation study,Ox-LDL and hepcidin were positively correlated with the Gensini score(r=0.350,0.376),which the correlation P was less than 0.05.5.The plasma levels of hepcidin in the group with CHD was positively correlated with the increase of Ox-LDL(r=0.177,P<0.05).The plasma levels of HDL and apoA1 were negatively correlated with the increase of hepcidin(r=-0.195,-0.191),which the correlation P was less than 0.05.Conclusion:1.The plasma levels of hepcidin in CHD,which was positively correlated with the severity of coronary atherosclerosis,may be one of the risk factors for CHD.2.2.The plasma levels of hepcidinin CHD,which was positively correlated with Ox-LDL and negatively correlated with HDL and apoAl,may affect the fat metabolism.3.Hepcidin may influence atherosclerosis by catalyzing the information of Ox-LDL.4.The plasma levels of hepcidin,which was markedly difference between the premenopausal and postmenopausal women,may affect the incidence of CHD in premenopausal and postmenopausal women.
