| Research objectives:High-fat diet does not only lead to metabolic diseases such as obesity,diabetes,hyperlipidemia,but also related to the cognitive function and emotional disorders.However,the mechanism of its influence has not been realized.Metabolic abnormalities induced by a high-fat diet in rodents can be used to mimic metabolic disorders in humans.In this study,C57bl/6 male mice were recorded the behavior changes after a high-fat diet.And then the effect of probucol on lipid metabolism and behavior changes should also be evaluated,so as to provide a new treatment idea for patients with abnormal lipid metabolism combined with cognitive disorder.Research Methods:Thirty-six C57bl/6 male mice were randomly divided into three groups after adaptive feeding for a week.Mice in control group were fed a normal diet,while the other two groups were fed a high-fat diet.After 5 weeks,blood glucose and lipid levels of the three groups were measured.Then the drug group was given the probucol containing in water(50 mg/L),the control group and the high-fat diet group fed water.After the intervention,the changes of lipid metabolism and behavior of mice in three groups were tested.Results:1.After 5 weeks of high-fat diet,the body weight,glucose and serum lipid level were significantly increased in mice.And the mice showed impaired spatial memory and anxiety-like mood;After 14 weeks on a high-fat diet,the mice developed a depressive state.2.After 12 weeks of intervention,probucol had no effect on body weight,blood glucose,and serum lipid level of mice fed on high-fat diet.However,probucol could improved cognitive impairment induced by the high-fat diet.Conclusion:1.High-fat diet are not only prone to metabolic diseases such as obesity,diabetes,hyperlipidemia,but also result in cognitive impairment,anxiety and depression in mice.2.Probucol can improve the cognitive impairment in mice induced by high-fat diet,but it have no effect on emotion disorder. |
