| Fici Hirtae Radix is the desiccation root of the Ficus hirta Vahl plant of the genus Moraceae.Traditional Chinese medicine holds that the Fici Hirtae Radix has the functions of invigorating spleen and tonifying qi and stimulating the circulation of the blood and causing the muscles and joints to relax.Modern studies suggest that Fici Hirtae Radix has excellent liver protection against binding stress and chemical liver injury,while it is unclear whether alcohol-induced liver injury is equally protective.In this study,the alcoholic liver injury model was established by alcohol gradient gavage mice to study the effect of Fici Hirtae Radix extract on the indexes of oxidative stress and inflammatory reaction in serum and liver were studied,and to study the expression of antioxidant protein and related proteins of ethanol metabolism pathway in liver,to explore the protective effect of Fici Hirtae Radix on alcoholic liver injury.The main research methods and results are as follows:(1)The mice were randomly divided into 9 groups according to the blank group,model group,Fici Hirtae Radix water and alcohol extraction group with low,medium and high doses.With the exception of the blank group,the mice in the other groups were made by gradient alcohol gavage with 42 degrees liquor,while the medication administration team was carried out with traditional Chinese medicine intervention.The positive group was intervened with 0.53 g/kg/d dose of flos puerariae,and the low,medium and high dose of Fici Hirtae Radix water and alcohol extraction groups were intervened with 0.5,1 and 2 g/kg/d.The molding period is 6 weeks.After the end of the experimental period,the liver index of each group was compared,and the same part of the liver of each group was observed histopathologically.The results showed that after 6 weeks of alcohol gavage,the liver index of the model group increased significantly,and the liver index of each medication administration team decreased compared with the model group.The liver index of high dose group of water and alcohol extraction from Fici Hirtae Radix was significantly lower than that of model group(P<0.05).Compared with the blank group,the liver cells in the model group were swollen,arranged and scattered,and the small degeneration around the central vein was significant.Compared with the model group,liver necrosis and inflammatory cell infiltration decreased in the Fici Hirtae Radix intervention group.(2)Blood samples were obtained by eyeball extraction and biochemical method was used to determine ALT、AST activity in serum.The results showed that compared with the blank group,the ALT(P<0.01)、AST(P<0.05)activity of the model group increased significantly,and compared with the model group,the ALT、AST activity of the intervention of Fici Hirtae Radix decreased to varying degrees,indicating that the Fici Hirtae Radix can reduce the liver injury and have liver protection effect.(3)The related indexes of ethanol metabolism pathway were mensurated.Biochemical method was used to determine ADH activity in serum.The expression level of CYP 2E1 protein in liver tissue was determined by Western blot.The results showed that the ADH activity and CYP 2E1 protein level in the model group were significantly higher than that in the blank group(P<0.05).Compared with the model group,the ADH activity and CYP 2E1 protein level in the intervention groups of Fici Hirtae Radix decreased to varying degrees,indicating that the liver protection of Fici Hirtae Radix may be related to maintaining the stability of liver cells,activating ADH activity in ethanol metabolism pathway,accelerating ethanol metabolism and reducing the expression of CYP 2E1 protein.(4)Determination of relevant oxidation indexes in liver tissue.Biochemical method was used to determine MDA、GSH content,GSH-Px and SOD activity in liver homogenate were determined respectively.The expression level of Nrf2、HO-1、SOD-1 protein in liver tissue was determined by Western blot.It turns out,Compared with the blank group,The MDA content of model group increased significantly(P<0.01),SOD activity decreased significantly(P<0.01),GSH content and GSH-Px activity decreased significantly(P<0.05);Compared with the model group,The MDA content decreased significantly(P<0.05),and the GSH content,SOD and GSH-Px activity were increased in the intervention groups of Fici Hirtae Radix to varying degrees.Meanwhile,Analysis of the results of related antioxidant proteins in the liver showed that the Nrf2、HO-1 and SOD-1 protein levels of model group more decreased significantly than blank group(P<0.05);Compared with the model group,Nrf2、HO-1、SOD-1 levels in the intervention groups of Fici Hirtae Radix were increased in different degrees.The results showes that Fici Hirtae Radix can reduce the content MDA lipid peroxidation products,increase the activity of SOD、GSH and GSH-Px,increase the expression of Nrf2 and its downstream related antioxidant protein HO-1、SOD-1,thus play its antioxidant role,reduce the damage of free radicals to the body and protect the liver.(5)Determination of relevant inflammatory indexes in liver tissue.The NF-κB、TNF-α、IL-1β and IL-6 levels in liver tissue were detected by ELISA method.The results showed that the NF-κB(P<0.01)、TNF-α、IL-1β and IL-6 levels in the model group were significantly higher than those in the blank group(P<0.05),and with the model group,the NF-κB、TNFα、IL-1β and IL-6 levels in the intervention groups of Fici Hirtae Radix decreased to varying degrees,indicating that Fici Hirtae Radix can reduce the level of NF-κB、TNF-α、IL-1β and IL-6 inflammatory factors in alcoholic liver injury and protect the liver. |
