| Objective To investigate the effects of gynecological Qing granules on inflammatory factors IL-1 β,IL-2,IL-6 in serum and TLR2/4-NF-κ B signaling pathway in uterine tissues of rats with pelvic inflammatory disease model.Methods Ninety female SD rats were adaptively fed for 5 d and randomly divided into normal group,model group,gynecological Qing granules low,medium and high dose group,and gynecological Qianjin capsule group.Except for the normal group,the pelvic inflammatory disease model was prepared by mechanical injury combined with phenol plasma in all groups.The rats were dissected under anesthesia after the last dose and the serum levels of IL-1β,IL-2 and IL-6 were measured by ELISA and the protein expression of TLR2,TLR4 and P65 in uterine tissues were detected by Western Blot.Results 1.Effects on the morphology of rat uterine tissuesThe morphological effects on the uterine tissues of rats were observed by visual and microscopic observation,which showed that the uterine tissues of the model group were blurred,some tissues were necrotic,and obvious congestion and edema,as well as local inflammatory cell infiltration and vacuolation-like changes were seen;while the uterus of rats in each treatment group had different degrees of improvement,among which the improvement was most obvious in the high-dose group of Gynecological Qing granules and Gynecological Qianjin capsule group.2.Effects on serum levels of IL-1β,IL-2 and IL-6 in ratsThe levels of IL-1 β and IL-6 in the model group were higher than those in the normal group,which was statistically significant(P<0.05),and the levels of IL-2 in the model group were lower than those in the normal group,indicating that the modeling was successful;after treatment,the levels of IL-1 β in the gynecological Qianjin capsule group were higher than those in the model group,and the levels of IL-1β in both the medium-dose and low-dose treatment groups of gynecologucal Qing granules were lower than those in the model group,which was statistically signifucant(P<The level of IL-2 in the model group was lower than that in the normal group,except for the low-dose group,the level of IL-2 in all the treatment groups was higher than that in the model group(P<0.05),and the difference was statistically significant,with the level in the high-dose group being the highest.The level of IL-6 in the model group was higher than that in the normal group,except for the low-dose group of Gynostemma,the level of IL-6 in all the treatment groups was lower than that in the model group(P<0.05),and the difference was statistically significant.3.Effects on the expression of VEGF and TGF-β1 in the uterine tissues of rats with pelvic inflammatory diseaseCompared with the normal group,the level of VEGF in the model group was lower than that in the normal group(P<0.05),which was statistically significant.Compared with the normal group,the level of TGF-β1 in the model group was lower than that in the normal group(P<0.05),and compared with the model group,the level of TGFβ1 in all treatment groups was higher than that in the model group(P<0.05),except for the low-dose group of Gynecomastia Qing granules(P>0.05),which was statistically significant.4.Effects on the expression of TLR2/4-NF-κ B signaling pathway-related proteins in rat uterine tissuesCompared with the normal group,the protein expression levels of TLR2,TLR4 and p65 in the uterine tissues of rats in the model group were significantly up-regulated;after treatment with gynecological Qing granules and gynecological Qianjin capsule,the protein expression levels of TLR2,TLR4 and p65 were significantly down-regulated,and the trend of down-regulation of TLR2,TLR4 and p65 was more obvious with the increase of the dose of gynecological Qing granules.The down-regulation effect was the most significant in the high dose group of Gynecomastia Qing granules(P<0.05).The results indicated that Gynecomastia Qing granules could inhibit the expression level of NF-κB signaling pathway-related proteins.Conclusion:Gynecomastia Qing granules can significantly improve the inflammatory responses such as uterine congestion and edema and inflammatory cell infiltration in rats with pelvic inflammatory disease model,and regulate the levels of inflammatory factors IL-1 β,IL-2 and IL-6 in the serum of rats with pelvic inflammatory disease model.The mechanism of Gynozyme Qing granules in the treatment of pelvic inflammatory disease may be achieved by regulating the protein expression of TLR2/4-NF-K B signaling pathway,inhibiting the expression of related inflammatory factors and thus reducing the response at the inflammation site. |
