| Obesity is a chronic metabolic disease with multiple factors interacting,and it is the inducement of fatty liver,diabetes and various cancer diseases.The dialectical and treatment of obesity in TCM has a long history,with a wide variety of drugs and complex action pathways.However,at the level of gene targets and action pathways,the treatment law of TCM for obesity is not yet clear,and there is a lack of research on syndrome differentiation and treatment.Therefore,based on the theory of network pharmacology,from the perspectives of ancient Chinese medicine books and clinical data,at the three levels of core targets,biological processes and action pathways,To study the TCM syndrome differentiation and treatment of obesity and the rules of drug use,to provide theoretical basis for the study of TCM clinical treatment of obesity.This research is divided into two parts.The first part is based on the network pharmacology analysis of the treatment prescriptions for the five diseases of obesity based on internal medicine of traditional Chinese medicine.The traditional Chinese medicine targets were obtained through the TCMSP database,the obesity targets were obtained through the Gene Cards database,and the intersection was calculated;the PPI protein interaction map between prescriptions and obesity was constructed in the String database;GO enrichment and KEGG pathway enrichment were performed through the Metascape database,and the Research on the law of syndrome differentiation and treatment based on the results of the five prescriptions.On the basis of the first part,the second part demonstrates the law of TCM syndrome differentiation and treatment through the potential prescriptions of clinical medical records.Through data mining,potential prescriptions are obtained from clinical medical records,and through network pharmacology analysis,the drug law of TCM syndrome differentiation and treatment,and the rule of monarch,minister and assistant are demonstrated at the three levels of core targets,biological processes and action pathways.The results of the study are as follows:1.The law of syndrome differentiation and treatment: the five groups of prescriptions all contain multiple core targets such as MAPK3 and JUN at the level of gene targets;at the level of biological processes,they all act on the response of cells to lipids,oxidoreductase activity and other processes;At the pathway level,Lipid and atherosclerosis,AGE-RAGE signaling pathway in diabetic complications and other pathways are used to regulate the absorption and decomposition of lipids and blood sugar,and control processes such as cell growth and apoptosis.2.Clinical data analysis results: Apriori analysis was used to obtain multiple groups of triple and quadruple prescriptions from clinical data,and 5 groups of clustered prescriptions were obtained through systematic cluster analysis.Pharmacological analysis of the combined formula network of 6 high-frequency traditional Chinese medicines: The prescription contains 215 targets for the treatment of obesity,including multiple core targets of JUN and TP53;at the level of biological processes,they acts on biological processes such as cell response to lipids,oxidoreductase activity,etc;regulates blood glucose and blood lipids through pathways such as Lipid and atherosclerosis,AGE-RAGE signaling pathway in diabetic complications,IL-17 signaling pathway,Regulation of lipolysis in adipocytes and has certain therapeutic effects in cancer pathways,Chemical carcinogenesis-receptor activation and other pathways,in line with The law of syndrome differentiation and treatment found in this paper. |
