| Objective:To study the serum metabolomics of Jian-Pi-Tiaogan Decoction in the treatment of simple obesity by liquid chromatography-mass spectrometry,screen out potential differential metabolites and metabolic pathways,and explore the mechanism of Jian-Pi-Tiaogan Decoction in the treatment of simple obesity.Methods:1 Thirty six C57 BL / 6J mice were divided into blank group and model group: after successful modeling,the blank group and model group were given normal saline,the western medicine group was given orlistat,and the traditional Chinese medicine group was given Jian-Pi-Tiaogan decoction.The weight of mice was recorded every week.After the 8th week,the contents of fasting blood glucose,triglyceride(TG),serum total cholesterol(TC),high density lipoprotein cholesterol(HDL-C)and low density lipoprotein cholesterol(LDL-C)were measured.2.The serum samples of mice in each group were detected by ultra-high performance liquid chromatography tandem quadrupole time of flight mass spectrometry(UPLC-Q-TOF/MS),and then the metabolic markers were analyzed and identified by principal component analysis(PCA)and orthogonal partial least squares analysis(OPLS-DA).The metabolic pathway enrichment analysis and metabolic pathway analysis of differential metabolites were carried out based on KEGG database.To explore the metabolic mechanism of obesity mice induced by high-fat diet and the intervention mechanism of Jian-Pi-Tiaogan decoction.Results: 1.Jian-Pi-Tiaogan Decoction and orlistat can reduce the weight of simple obese mice,and the effect of Jian-Pi-Tiaogan decoction is likely to be better than orlistat.In the aspect of Lee’s index,compared with that before treatment,the Lee’s index in the traditional Chinese medicine group decreased significantly(P < 0.01),and there was significant difference in the western medicine group(P < 0.05).It is suggested that the weight loss effect of traditional Chinese medicine group may be better than that of orlistat group,and Jian-Pi-Tiaogan Decoction and orlistat are very likely to effectively reduce the Lee’s index of mice.Orlistat can reduce fasting blood glucose in obese mice;In terms of lipid metabolism,both drugs could reduce the levels of TG and TC in simple obese mice(P < 0.01),and the curative effect of Jian-Pi-Tiaogan decoction was better than orlistat(P < 0.05);For LDL-C,compared with the model group,there was significant difference between the traditional Chinese medicine group(P < 0.01)and the western medicine group(P < 0.05).For HDL-C,compared with the model group,it decreased in the traditional Chinese medicine group and the western medicine group after treatment,but there was no significant difference;It can be seen that Jian-Pi-Tiaogan Decoction and orlistat have a good effect on improving glucose and lipid metabolism,and the curative effect of Jian-Pi-Tiaogan decoction may be better than orlistat in TG and TC indexes.2.Based on the analysis of LC-MS technology,75 kinds of foreign bodies with poor metabolism related to the pathology of simple obesity were identified,of which 23 metabolites were up-regulated and 52 metabolites were down regulated;There were 24 different metabolites in the traditional Chinese medicine group compared with the model group,of which 15 metabolites were up-regulated and 9 metabolites were down regulated;There were 34 different metabolites in the serum between the model group and the western medicine group,of which 22 metabolites were up-regulated and 12 metabolites were down regulated.There are two significant metabolic pathways in the model group and the traditional Chinese medicine group: linoleic acid metabolism and vitamin B6 metabolism;There are two significant in model group and Western medicine group: purine metabolism and sphingolipid metabolism;There were 9significant in the model group and the blank group,which were glycerol phospholipid metabolism,choline metabolism in cancer,retrograde endocannabinoid signaling,autophagy-other,Kaposi sarcoma-associated herpesvirus infection,autophagy-animal,necroptosis,sphingolipid signal pathway,Glycosylphosphatidylinositol(GPI)-anchor.Conclusion:1.Jian-Pi-Tiaogan can effectively reduce the weight of simple obese mice and improve the disorder of glucose and lipid metabolism.2.The metabolic pathway of vitamin B and linoleic acid in liver can be regulated by vitamin B.To provide strong biological support for the study of simple obesity. |
