Objective:To investigate the effects of sevoflurane postconditioning on cognition function and hippocampal inflammation in myocardial ischemia reperfusion rats.Methods:Thirty-six male SD rats aged 2~3 months and weighing 200~250g were randomly divided into 3 groups: Sham operation group(Sham group),myocardial ischemia reperfusion group(MI/R group),sevoflurane postconditioning + myocardial ischemia reperfusion group(SP group).Myocardial ischemia-reperfusion model was prepared by30 min occlusion of left anterior descending branch of left anterior descending coronary artery,and rats in SP group inhaled 2.4% sevoflurane for 15 min from 10 min before reperfusion.After 120 min of reperfusion,the serum cTnI concentration was detected by ELISA.Morris water maze test was used to evaluate the cognitive function of rats at 1-5d after reperfusion.After the water maze experiment,the hippocampal tissue of rats was collected,and then the apoptosis index of hippocampal neurons was detected by TUNEL method.The contents of IL-1β,IL-6 and TNF-α in hippocampal tissues were detected by ELISA method.The expression levels of IL-1β,IL-6 and TNF-α protein in hippocampal tissues of rats were detected by Western blot.Results:Compared with Sham group,MI/R group and SP group had higher serum cTnI concentration(P<0.05),longer escape latency(P<0.05),shorter stay time in the quadrant of the original platform(P<0.05),fewer times of crossing the original platform(P<0.05),increased levels and protein expression of IL-1β,IL-6,TNF-α(P<0.05)and increased hippocampal neuron apoptosis index(P<0.05);Compared with MI/R group,SP group had lower serum cTnI concentration(P<0.05),shorter escape latency(P<0.05),longer stay time in the quadrant of the original platform(P<0.05),more times of crossing the original platform(P<0.05),decreased levels and protein expression of IL-1β,IL-6,TNF-α(P<0.05)and decreased hippocampal neuron apoptosis index(P<0.05).Conclusion:Sevoflurane postconditioning can improve the cognitive function of myocardial ischemia-reperfusion rats,and the mechanism may be related to the decrease of the content and protein expression of IL-1β,IL-6 and TNF-α in hippocampal tissue,and the decrease of neuronal apoptosis.
