Differential Analysis Of The DKK1 Expression Levels In The Different Grades Of The Meningioma | | Posted on:2023-05-14 | Degree:Master | Type:Thesis | | Country:China | Candidate:J Y Xu | Full Text:PDF | | GTID:2544306818450694 | Subject:Surgery | | Abstract/Summary: | PDF Full Text Request | | Objective: By measuring the expression levels of DKK1 at the protein and gene levels in meningioma tissue,the expression differences of DKK1 in different pathological grades of meningioma were explored and analyzed.Methods: Thirty patients with meningioma admitted to the Department of Neurosurgery of the Second Hospital of Hebei Medical University from March 2021 to January 2022 were selected.Another 8 patients who underwent decompressive craniectomy due to craniocerebral trauma were collected as controls.The meningioma tissue samples in this study were graded using the World Health Organization(WHO)grading criteria for central nervous system(CNS)tumors(2016 edition),including 19 WHO grade I benign meningiomas and 8 WHO grade II atypical meningiomas 3 cases of WHO grade III anaplastic(malignant)meningioma.All patients underwent Simpson grade I or II surgical resection,and the postoperative pathological diagnosis results were confirmed by 2 or more experienced pathologists in the Second Hospital of Hebei Medical University.Meningioma patients were all first cases without other complications.The control group consisted of 8 patients undergoing decompressive craniectomy due to craniocerebral trauma who underwent decompressive resection of brain tissue during the same period.Within 10 minutes of taking the specimens,some specimens were placed in a liquid nitrogen tank for freezing for future use,and some specimens were placed in a paraformaldehyde solution for storage.According to the grade of meningioma and the control group,a total of four groups were tested.The expression levels of DKK1 in meningioma tissue and normal brain tissue were detected by immunohistochemical staining and RT-PCR from protein and gene levels.Results: 1.Immunohistochemical test results.The expression of DKK1 in meningioma tissue was higher than that in normal brain tissue,and the pigmented granules were concentrated in the cell membrane and cytoplasm.The AOD values of the control group and the meningioma groups of all grades(WHO grades I-III)were compared between the groups,and there were significant differences in the expression of DKK1 between the groups,with statistical significance(P<0.001).2.RT-PCR test results.The expression of DKK1 gene m RNA in all grades of meningioma tissues was higher than that in the control group,and there were significant differences in the expression levels among the groups.Conclusions:1.The expression of DKK1 in all grades of meningioma is higher than that in normal brain tissue.(P<0.01)2.The expression of DKK1 was positively correlated with the pathological grade of meningioma. | | Keywords/Search Tags: | Meningioma, DKK1, DKK family, Immunohistochemistry, RT-PCR | PDF Full Text Request | Related items |
