| Theory of“simultaneous treatment of lung and intestine”(STLI)is one of the original concepts for fighting ailments in practice of traditional Chinese medicine(TCM).As the classical‘Xuanbai Chengqi decoction”(XCD),the prescription of STLI has outstanding clinical advantages in treating refractory diseases of the lung and intestine,but its effective substances and mechanism are still unclear.Based on the specific relationships betweenβ2/M3/Cys LT1receptors and asthma treatment,this study investigated the anti-asthmatic effects of XCD using the rat asthma model,and analyzed the roles of the prescription and its separated"treating lung"(TL)and"treating intestine"(TI)recipes in regulatingβ2/M3/Cys LT1receptor expressions.With the utilization of immobilized receptor chromatography,further work was performed to recognize the targeting compounds.The accomplishment of this proposal would clearly identify the target receptors and the functional substances of XCD of STLI for fighting asthma.It is also promising to elucidate preliminarily the scientific meanings of the compatibility of XCD.The full text is divided into four chapters,and the main contents are as follows:1.The therapeutic effects of different dose of XCD were assessed with rat asthma model.The model was established by ovalbumin(OVA)sensitization and inhalation.Several aspects including behavior symptoms,inflammatory levels and tissue histopathology were applied for the evaluation.The results showed that XCD significantly relieved the symptoms of asthma rats,reduced the levels of Ig E,OVA-Ig E and the number of inflammatory cells in both serum and bronchoalveolar lavage fluid(BALF)of asthmatic rats,and improved the lung pathological changes in asthmatic rats.The effects of high dose group were most obvious.The above results indicated that XCD has the action of fighting asthma.2.The pharmacodynamic features of XCD,TL and TI recipe were examined with medium dose treatment,while theβ2/M3/Cys LT1receptor expressions in the rats lung tissue were detected through immunoblotting and immunohistochemistry methods.The results showed that the anti-asthmatic effects of XCD were better that of TL and TI recipes.The XCD,TL and TI could increaseβ2receptor expression,inhibit expressions of M3and Cys LT1receptor in the rats lung tissue.The results suggested that XCD,TL and TI recipe all could excert influences on theβ2/M3/Cys LT1receptor,which would lead to the synergistic effects of XCD of STLI in treating asthma.3.With the utilization of immobilizedβ2and M3receptor chromatography,further work was performed to recognize theβ2and M3receptor targeting compounds in XCD,TL and TI recipe.The compounds of interesting were collected and identified by HPLC-MS.The results showed that amygdalin was the active compound targeting bothβ2and M3receptor in TL recipe.In TI recipe,catechins,rhein and emodin were the active compounds targetingβ2receptor,as well as emodin was the active compound of M3receptor.Moreover,the screening compounds in XCD were same as those of TI and TL recipe.It is concluded that these substances may be the material basis of XCD of STLI in treating anthma. |
