| Since the discovery of the novel coronavirus in Wuhan at the end of 2019,there have been more than 440 million confirmed cases of novel coronavirus pneumonia worldwide and more than 6.02 million deaths.China has effectively controlled the spread and spread of the epidemic through dynamic clearing and large-scale popularization of vaccines.However,due to the continuous development of the novel coronavirus,the domestic epidemic has shown local high-aggregation and scattered outbreaks,leading to the problem of medical jams in a short period of time.At the same time,the secretion of SARS-CoV-2neutralizing antibodies after vaccination is also one of the key factors in preventing the spread of the virus.Therefore,for the two problems of rapid detection of SARS-CoV-2virus and improvement of methods for evaluating the efficacy of vaccination,during the postgraduate period,I was committed to the rapid,accurate and stable detection of SARSCOV-2 antigen and the accurate and accurate detection of the efficacy of SARS-CoV-2neutralizing antibody,so as to effectively reduce the medical pressure of the centralized detection of SARS-COV-2.The main contents are as follows:1.Establishment of a latex microsphere-based immunochromatographic method for the detection of SARS-CoV-2 antigensIn order to meet the demand for rapid detection of SARS-CoV-2 antigens,this paper uses latex microspheres as the carrier and NHS-EDC as the "bridge" to connect the proteins to realize the chemical adsorption of chemical bonds to surface proteins.Through the principle of double-antibody sandwich detection antigen,the established immunosensors realizes the rapid,accurate and low-cost detection of SARS-CoV-2 antigen.Under the optimal conditions,the specificity,anti-interference,repeatability,stability and sensitivity tests of the immunosensor showed excellent experimental results.To evaluate the clinical applicability of the proposed immunochromatographic sensor,659 samples(230 positive and 429 negative samples)were tested using immunochromatographic sensor and r RT-PCR tests at the same time,and the Youden index for this clinical test was 0.9615.It shows that the immunochromatographic sensing can provide a good auxiliary means for grassroots personnel to realize the early self-examination of the new coronavirus and prevent the further spread of the epidemic.2.Establishment of a method for the detection of SARS-CoV-2 neutralizing antibodies based on a storable chemiluminescent freeze-dried mixtureIn order to achieve an effective evaluation of the production of SARS-CoV-2neutralizing antibodies in the human body after injection of the novel coronavirus vaccine,a chemiluminescence freeze-dried mixture that can be used for storage at room temperature was prepared by freeze-drying.By optimizing the formulation of FC buffer,the coated protein can maintain its biological activity and stability.Under the optimal detection conditions,its accuracy,precision and stability were tested by immunocompetitive method.The results showed that the coefficient of variation was only 1.93% and the recovery rate was 102.20%.At the same time,the mixture could be stored at room temperature for a period of time.Compared with traditional cold chain transport storage,the freeze-dried product also has higher accuracy,precision and stability.The proposed freeze-dried mixture is easy to use,store and transport conveniently,and has high accuracy,which has great clinical application for the protection evaluation of SARS-CoV-2 neutralizing antibodies. |
