| Objective: This study intends to clarify the expression pattern of Lonp2 in HNSCC and further explore the function and mechanism of Lonp2 in regulating metastasis of human head and neck squamous cell carcinoma,and provide new strong clues for clinical treatment.Methods: 1.Explore the specific function of Lonp2 knockdown/overexpression in HNSCC and its influence on the occurrence and development of HNSCC: 1)Prepare Lonp2 knockdown/overexpression lentivirus vectors of TU686(human laryngeal cancer cell line)and Fadu(human hypopharyngeal cancer cell line)respectively,and set 2 interference targets in the knockdown group;2)TU686(human laryngeal cancer cell line)and Fadu(human hypopharyngeal cancer cell line)with stable knockdown/overexpression of Lonp2 were constructed and the effects of knockdown/overexpression of Lonp2 on the growth,invasion and metastasis of HNSCC tumor cells were detected by cell scratching test,cell invasion and migration test.The effect of Lonp2 on the development of HNSCC was preliminarily investigated at the cellular level.2.Western blot was used to detect the effect of Lonp2 on the expression of Snail,Twist and Vivimentin,and further analyze the effect and mechanism of Lonp2 on distant metastasis of HNSCC.Results:(1)TU686(human laryngeal cancer cell line)and Fadu(human hypopharyngeal cancer cell line)with Lonp2 knockdown/overexpression were successfully constructed in the early stage.After screening out TU686 and Fadu cell lines with Lonp2 high expression,it was found that the cell proliferation capacity of the two HNSCC cell lines was enhanced.(2)In this study,it was found that the migration and invasion ability of two HNSCC cell lines(TU686 and Fadu)were enhanced after Lonp2 overexpression,and the migration and invasion ability of HNSCC cell lines were decreased after Lonp2 knockdown.(3)Lonp2 can promote epithelial-mesenchymal transition(EMT)in HNSCC cells.Overexpression of Lonp2 increased the expression of Snail,Twist(Twist protein)and VIM(vimentin)related transcription factors affecting the EMT pathway,and enhanced the invasion and metastasis ability of HNSCC,while knockdown of Lonp2 significantly reduced the expression of Snail,Twist(Twist protein)and VIM(vimentin)compared with the control group.Therefore,this study concluded that Lonp2 can induce the EMT process of HNSCC by regulating Snail,Twist and VIM,thus participating in the occurrence and development of head and neck tumors.Conclusion:The increased expression of Lonp2 in HNSCC tissue can functionally promote the proliferation of HNSCC cells in vitro,and it may induce the upper cortical cell-mesenchymal transformation process to promote the migration and invasion of HNSCC by up-regulating Snail,Twist and VIM. |
