| Objectives: To explore the basic characteristics of gut microbiota composition in patients with Hashimoto’s thyroiditis,and to find out the bacteria significantly related to hashimoto’s thyroiditis,in order to provide new research ideas for the treatment of hashimoto’s thyroiditis.Methods: A total of 25 stool samples from hashimoto’s thyroiditis patients who met the inclusion and exclusion criteria in the Outpatient department of Breast and Thyroid of Qinghai University Affiliated Hospital from October 2020 to March 2021 were collected,which were HT group.At the same time,20 stool samples from healthy volunteers in the physical examination center of our hospital during the same period were enrolled in the HC group.After passing the quality inspection,DNA extraction and PCR amplification were performed for gut microbiota of the two groups,and V3-V4 region of 16 Sr RNA gene was sequenced.The sequencing results of the two groups were evaluated,and OTU clustering,diversity analysis,species composition characteristics and bioinformation analysis of species differences were carried out.The gut microbiota function was predicted using KEGG database,and the differences of microbiota function were compared.Results: 1.Sequencing analysis showed that the two groups obtained a total of2397814 high-quality effective sequences,with an average of(53285±4838)effective sequences generated per sample,indicating sufficient amount of sequencing data;2.OTU clustering results showed that the two groups obtained a total of 851 OTUs,the HT group had 169 unique OTUs,the HC group had 107 unique OTUs;3.Diversity analysis showed that there was no significant difference in Alpha diversity between the two groups(P>0.05),but the weighted unifrac and unweighted unifrac distance of Beta diversity between the two groups was significant difference(P<0.01);4.Comparison of species composition and differences between groups showed that: The species composition and abundance of phylum,class,order,family and genus were different in the two groups.LEf Se analysis showed that there were24 and 21 difference dominant microbiota with potential biomarkers in HT group and HC group(P<0.05).6 key difference bacteria were obtained by comparing the top 10 key species in abundance at each level,Compared with HC group,the abundance of Candidatus_Sacchariabacteria,Enterobacteriales and Enterobacteriaceae in HT group was significantly increased,the abundance of Deltaproteobacteria,Desulfovibrionales and Porphyromonadaceae was significantly decreased(P<0.05);5.The prediction analysis of gut microbiota abundance function between the two groups showed that there were statistically significant differences in gut microbiota function in 8 metabolic pathways(P<0.05),including bacterial invasion of epithelial cells,drug metabolism enzymes and phosphoptransferase system et.Coclusions: The gut microbiota composition of HT patients is significantly different from that of healthy people,and the significant changes in the abundance of different microbiota and the functional pathways of the microbiota may be related to the occurrence and development of HT.Differential microbiota may contribute to gut microecological therapy for HT patients. |
